摘  要:目的探讨脑缺血-再灌注损伤对大鼠脑组织髓磷脂相关糖蛋白(MAG)表达的影响。方法应用线栓法制作大鼠脑缺血-再灌注损伤模型;采用免疫荧光化学和免疫印迹法研究大鼠脑缺血-再灌注损伤后MAG表达的定位和定量的变化。结果脑缺血-再灌注后4 h和24 h大鼠皮质区MAG的表达有增高趋势,但是差异无统计学意义;基底核区MAG的表达4 h即开始升高,但差异亦无统计学意义。缺血组基底核区MAG 24 h的表达与无缺血组比较,差异有统计学意义,(P<0.05)。免疫荧光实验显示,阳性表达在基底核的向质区域,与细胞核的复染发现,主要表达在细胞间。结论大鼠腑缺血再灌注后MAG的表达在皮质区有增加趋势,在基底核区的表达明显增高,24 h缺血组显著高于无缺血组。提示脑缺血-再灌注损伤不仅损伤神经元,其白质也存在损伤,干预白质损伤可能是脑缺血-再灌注损伤新的治疗靶点。Objective To investigate the influence of cerebral isehemia-repeffusion injury on myelinassociated glycoprqtein (MAG) expression in rats. Methods A model of rat cerebral isehemia-repeffusion injury was induced by the intraluminal suture method. The immunofluoreseenee technique and Western blot were used to study the changes of localization and quantification of MAG expression after cerebral is- chemia-repeffusion injury in rats. Results There was an increased trend in the expression of MAG in cortical areas at 4 and 24 hours after ischemiaepeffusion in rat, but there was no statistical difference. The expression of MAC in basal ganglia areas began to increase at 4 hours, but there was no statistical difference. There was statistical significance for the expression of MAC in basal ganglia at 24 hours between the iscbemic group and the non-ischemic group (P 〈 0. 05). Immunofluorescenee experiments showed that the positive expression was in the white matter of basal ganglia, and the retained nuclei found mainly in the expressed cells. Conclusion There was an increased trend in the expression of MAG in cortical areas af- ter cerebral ischemia-repeffusion injury in rats, especially in basal ganglia, and at 24 hours the expression increased more significantly in the ischemie group than that in the non-isehemie group. It suggested that cerebral ischemia-reperfusion injury not only damage neurons, but also damage white matter. The intervention of white matter injury may be a new therapeutic target for isehemia-reperfusion injury.
