摘 要:利用DF-1细胞系、ELISA群特异性抗原检测以及亚群特异性间接免疫荧光法,从湖北省某鸡场送检的海兰褐蛋鸡病料中分离鉴定出1株J亚群禽白血病病毒(ALV-J),命名为HB09JY03。为了解其分子特征,对其进行了全基因组测序,并与其他毒株进行了比较。结果表明,HB09JY03分离株的gag和pol基因非常保守,与各参考毒株的同源性为97.5%~99.0%;env基因的同源性为88.4%~97.5%;其3′UTR出现部分rTM区段缺失现象。与参考毒株相比,HB09JY03分离株与HPRS-103株的亲缘关系较近,可作为我国蛋鸡ALV-J株生物学特性和致病机制研究的良好材料。HB09JY03,a strain of the subgroup J avian leukosis virus(ALV-J) ,was isolated from Hyline laying hens on a farm of Hebei Province in China,then it was identified by ELISA,PCR and indirect immunofluorescence assay with p27 antigen which is specific to ALV subgroup.The genome of the virus strain was sequenced and compared with that of other ALV-J strains.The results showed that the gag and pol genes of the isolate were highly conserved,sharing 97.5% to 99.0% identities with those of other ALV-J isolates.The env gene of the isolate shared 88.4% to 97.5% homology with that of the other ALV-J isolates,and had partial deletions of the rTM(redundant TM) in 3′UTR(untranslated region) .Overall,the HB09JY03 strain shared a higher similarity with the prototype ALV-J strain HPRS-103 than other domestic ALV-J isolates,which might provide an excellent material for researching biological characteristics and pathogenesis induced by ALV-J strain on egg-type chicken in China.
