摘  要:目的:研究Ada蛋白质对alkA基因表达的调节,探讨Ada蛋白质与alkA基因有效转录的关系。方法:采用DNA顺序测定和体外转录的方法,以lac基因启动子为对照进行实验。结果:Ada蛋白质加入以突变和野生alkA基因启动子为模板的体外转录体系中,产生的alkAmRNA高于无Ada蛋白质组;Ada蛋白质加入lac基因启动子转录体系中,产生的lacmRNA与未加Ada蛋白质组基本相同。结论:Ada蛋白质对lac基因启动子转录调节作用不明显,对于突变的和野生的alkA基因转录均有促进作用,这种调节作用可能发生在启动子上游区。Objective: We investigated the relationship between Ada protein and the effective transcription of alkA gene in order to study the regulation of alkA gene expression by Ada protein. Methods: DNA sequencing and in vitro transcription had been used and the promoter of lac gene had been used as the control. Results: The amount of alkA mRNA transcripted from mutant and wild alkA gene promoters was higher in the group in presence of Ada protein than that in the group without Ada protein. There wasn't apparent difference between the two groups with and without Ada protein when lac gene promoter was used as template. Conclusion:T5BZThe transcription of lac gene promoter wasn't regulated by Ada protein,but the transcription of mutant and wild alkA gene promoters were effectively upregulated by Ada protein. This regulatory effect might be on the upstream of alkA gene promoter.
