摘  要:目的检测MMP9在鼻咽癌细胞株及细胞株裸鼠成瘤瘤块组织中的表达,探讨MMP9的表达与鼻咽癌发生、发展的关系。方法采用细胞免疫组织化学技术、RT-PCR、Western blot检测鼻咽癌细胞系CNE1、CNE2、HNE1、HNE2和SUNE15株细胞株中MMP9表达;免疫组织化学(SABC法)检测HNE1和CNE2细胞裸鼠成瘤后瘤块MMP9的表达。结果5株鼻咽癌细胞株中,免疫组化显示HNE1和SUNE1细胞中MMP9蛋白表达阳性。RT-PCR及Western Blot检测结果显示,HNE1和SUNE1细胞中MMP9 mRNA及蛋白高表达。HNE1和CNE2细胞裸鼠种植成瘤后,5例瘤块中MMP9蛋白均强阳性表达。结论MMP9在鼻咽癌细胞系中表达阳性,成瘤后表达增强,提示MMP9的表达可能与鼻咽癌侵袭转移有关。ObjectiveTo detect the expression of MMP9 in nasopharyngeal carcinoma (NPC) cell lines and implanting tumors of NPC cell lines in nude mice, and to investigate its significance to the occurrence and development of NPC.MethodsUsing immunohistochemical method, RT-PCR and Western blotting, the expression of MMP9 in five NPC cell lines (CNE1, CNE2, HNE1, HNE2 and SUNE1) were screened. Immunohistochemical method (SABC method) was used to detect MMP9 expression in implanting tumors of HNE1 or CNE2 cells in nude mice.Results In five NPC cell lines, MMP9 protein could be detected in HNE1 and SUNE1 cells with immunohistochemical method. The strong mRNA and protein levels of MMP9 in HNE1 and SUNE1 cells were detected with RT-PCR and Western blot. The strong MMP9 expression was detected in implanting tumors of HNE1 or CNE2 cells in five nude mice.ConclusionThe expression of MMP9 is positive in NPC cell lines, and is stronger in the implanting tumors, which suggests that the expression of MMP9 may contribute to the invasion and metastasis of NPC.
