摘  要:目的通过研究3种类型的CpG-ODN对哮喘小鼠Th1/Th2相关细胞因子表达的影响,探讨CpG-ODN治疗支气管哮喘的可能机制。方法以鸡卵蛋白(OVA)免疫小鼠建立哮喘模型,CpG-ODN干预组分为C1、C2、C3组,分别给予3种CpG-ODN腹腔注射,观察小鼠肺组织病理改变,支气管肺泡灌洗液(BALF)中白细胞总数、嗜酸性粒细胞数的变化;ELISA法检测BALF及血清中IL-4和IFN-γ的含量变化。结果3个CpG-ODN干预组病理改变较模型组轻,但3组之间镜下观无差别;3组CpG-ODN干预后BALF中细胞总数和EOS计数及IL-4均低于哮喘组,C3组最低(P<0.01),C1组与C2组之间无明显差异(P>0.05);3组CpG-ODN干预后的IFN-γ浓度均高于B组,C3组最高,C1组高于C2组(P<0.01)。结论3种类型的CpG-ODN对哮喘小鼠气道炎症反应及Th1/Th2失衡均有较好的逆转作用,由于结构及活性特点,C型CpG-ODN较A、B型效果更好。【Objective】To explore the effect of three types CpG-ODN on the expression of Th1/Th2 cytokines in a model of asthma. 【Method】 Asthmatic mice models were established by ovalbumin intraperitoneal injection and atomization inhalation. Then the mice were intraperitoneally injected with three types CpG-ODN. The serum and BALF levels of Th1 cytokine IFN-γ and Th2 cytokine IL-4 were measured by ELISA. Meanwhile, bronchoalveolar lavage fluid were collected for counting total inflammatory cells and eosinophils. 【Results】In CpG-ODN treatment group, the pulmonary inflammation was remarkably relieved than that of model group, but there was no difference in three intervention groups. The level of IL-4 ,WBC and EOS count decreased in three intervention groups, C3 group was lowest(P 0.01), and there was no obviously discrepancy in C1 and C2 group. The level of IFN-γ was increased in three intervention groups, C3 group was highest(P 0.01), and C1 was higher than C2 group(P 0.01).【Conclusion】 Three types CpG-ODN all can relieved the pulmonary inflammation and correct the imbalance of Th1/Th2 on cytokine level in the model of asthma. Resulted in its construction features, C type CpG-ODN has more efficiency than A and B types.
