大豆苷原和染料木黄酮对小鼠子宫内膜癌TNF-α、IL-1α及IL-6表达的影响

摘　　要：目的探讨大豆苷原和染料木黄酮在雌二醇-17β(E2)所诱发的小鼠子宫内膜癌TNF-α、IL-1α、IL-6 mRNA表达变化中的作用。方法利用定量RT-PCR法检测小鼠子宫内膜癌细胞TNF-α、IL-1α、IL-6 mRNA的表达量。结果 E2单独投与群TNF-α、IL-1α、IL-6 mRNA分别为(0.605±0.011)、(2.796±1.569)、(2.809±0.011)。与E2单独投与群比较,E2和染料木黄酮和大豆苷原联合投与群,能够明显抑制E2刺激所诱发的TNF-α、IL-1α及IL-6表达的增强。结论 E2皮下注射的雌性ICR小鼠给予染料木黄酮和大豆苷原后,能够诱发TNF-α、IL-1α、IL-6 mRNA的表达下调,对子宫内膜癌的预防有一定的意义。【Objective】To study the effect of genistein and daidzein on the expression of TNF-a,IL-1a and IL-6 mRNA in estrogen（E2）-induced endometrial cancer in ICR mice,and to study it＇s preventive value to tumor.【Methods】Immunohistochemical method and semiquantitative RT-PCR technique were applied to detect the expression level of TNF-a,IL-1a and IL-6 mRNA in the endometrial cancer cells in ICR mice.【Results】When estrogen（E2） was given alone,the expression of TNF-a,IL-1a and IL-6 mRNA increased significantly.While estrogen（E2） and genistein or daidzein were given together,genistein or daidzein significantly decreased the expression of TNF-a,IL-1a and IL-6 induced by estrogen（E2）.【Conclusion】When the female ICR mice with subcutaneous injection of estrogen（E2） induced endometrial cancer are given genistein or daidzein,the expression of TNF-a,IL-1a and IL-6 mRNA decreases.It has certain effect on the prevention of endometrial cancer.
