摘 要:用抑制性消减杂交(SSH)方法研究动物杂种相关基因表达差异。选用北京鸭、法国番鸭及其正反交F_1代,以杂种组为处理,亲本组为对照,建立肌肉组织SSH基因文库,随机选取1 000个单克隆测序,并与GenBank序列进行比对分析。结果共发现40个新ESTs,93个未知功能蛋白基因,34个已知功能蛋白基因。在34个已知功能蛋白基因中,杂种表达上调的基因有23个,杂种表达下调的有11个。研究表明杂种优势分子遗传基础复杂,涉及能量代谢、信号传导及调控、转录调控、蛋白质合成加工等基因的差异表达。This experiment was conducted to study the expression difference of hybrid vigor relevant genes through the method of suppression subtractive hybridization(SSH).Beijing duck,France muscovy duck and the their mutual hybrids were divided into 2 groups,the hybrids being the treatment and the parents being the control to construct SSH gene library.1 000 clones were picked randomly for sequencing.The nucleotide sequences were analyzed through NCBI blasting procedure.The blasting results revealed 40 new ESTs,93 hypothetical protein transcripts and 34 functional protein gene,among which 23 were up-regulated and 11 were down-regulated in hybrids.These results indicated that the molecular basis of hybrid vigor refers to complicated progress including energy metabolism,signal transduction,transcript regulation and protein synthesis and posttranslational processing.
