摘  要:目的:了解信号转导和转录活化因子3(STAT3)在新生大鼠未成熟肺高氧损伤过程中的表达规律及其与早产儿慢性肺疾病的内在联系。方法:取22 d胎龄足月新生SD大鼠,随机分为实验组(高氧组)与对照组(空气组),每组各8只大鼠,实验组生后立即置入体积分数>95%持续氧环境中饲养;对照组则在空气中饲养。两组新生大鼠于生后第3、7、14天随机抽取后处死并取其肺组织,HE染色观察肺组织病理变化,Masson三色染色判断肺组织纤维化程度,免疫组化测蛋白含量,逆转录和聚合酶链反应(RT-PCR)方法检测肺组织STAT3 mRNA的表达。结果:空气组生后第3、7、14天各亚组未见肺损伤的病理性改变,也没有胶原沉积增多现象;高氧组生后3 d肺泡内炎症反应及胶原纤维的沉积均不明显;第7天时可见大量炎细胞浸润,胶原沉积开始增多;第14天时炎细胞浸润减少,纤维化程度更加显著。第3、7、14天高氧组STAT3在肺泡上皮细胞、支气管上皮细胞、肺血管内皮细胞及炎症细胞等的表达明显增强,与同时期的空气组相比均显著增高,在高氧组中,以第7天时表达最强。第7、14天高氧组中肺组织的STAT3 mRNA的相对表达量明显高于对照组,其中以第7天高氧组的相对表达量最高,而两组的第3天亚组并未见明显差别。结论:高氧可造成新生鼠肺组织明显的病理损伤,早期为炎性反应阶段,后期纤维化程度逐渐增强。高氧暴露早期STAT3在肺组织中的表达随着暴露时间的延长而增加,提示STAT3主要在早期的急性炎症阶段起作用,在后期纤维化阶段呈逐渐降低趋势,未证实其与高氧肺损伤后期的纤维化有相关性。Aim:To investigate signal transducer and activator of transcription 3(STAT3) expression in the lung of neonatal premature rats exposed to hyperoxia,and to explore the internal relationship between STAT3 and chronic lung disease in preterm infants. Methods: Neonatal Sprague-Dawley rats of 22-day gestational age were randomized into experimental group and control group,the neonatal rats in experimental group were fed in high concentration of oxygen(inhaled 95% oxygen) and the control groups were fed in the room air.According to the number random method eight neonatal SD rats were executed at postnatal 3rd,7th and 14th day respectively.Then the lung tissues were taken for observing,the pathological changes with HE stain,Masson trichrome stain to assess the interstitial fibrosis severity of lung.The expression of STAT3 in lung tissues were analyzed by immunohistochemical techniques.And also examined the expression of STAT3 messenger ribonucleic acid(mRNA) in rat lung by fluorescent real-time RT-PCR.Results: Pathological change were not seen and collagen fibers did not increase in air group at postnatal 3th,7th and 14th day.In hyperoxic group at postnata 3th,alveolar inflammation and deposition of collagen fibers were not obvious;A large number of inflammatory cell infiltration were showed on 7th days,collagen deposition began to increase;14th days inflammatory cell infiltration reduced,fibrosis was more significant.In hyperoxic group,The expression of STAT3 in alveolar and bronchi epithelial cells and vascular endothelial cells as well as inflammatory cells were stronger on the postnatal 3th,7th and 14th day in hyperoxic group,the difference was significant compared with air group.The expression of STAT3mRNA in the lung of both groups at 3th were similar,by fluorescent real-time RT-PCR technique.Conclusion:Hyperoxic exposure can cause lung damage,inflammatory response is seen at early stage and the degree of fibrosis were increase gradually,furthermore along with the extension of oxygen uptake tim
