人肝癌组织中芳香基酰胺酶表达与异常改变的研究

摘  要:为探讨肝癌不同部分组织中AAD的表达和改变规律。方法将自身配对的肝癌、癌周和远癌组织中总RNA和AAD纯化,并分析了肝匀浆中AAD及不同分子形式AAD比活性,并与对应组织中GGT进行了比较。结果肝癌组织总RNA浓度,癌组织明显低于癌周和远癌组织(P<0.01);总AAD比活性(U/g)在癌组织明显高于远癌组织(P<0.05);肝癌组织中膜结合性AAD升高,但差别不明显(P>0.05),而可溶性AAD在肝癌组织中明显地高于癌周和远癌组织(P<0.05)。结论资料提示肝癌组织可表达大量AAD,且以可溶性AAD升高为主。Aim To explore the expression and alteration of eqlamidase (AAD) in different pale of hepatoma tissues.Methods The loud RNA and AAD were Purified, the specific activities of total AAD and different molecular forms of AADs inthe tissue homogenates were investigated and compared with the corresponding GGT in self-paired hepatoma tissues, adjacentcancers and distal cancers. Results The total RNA concentration in hepatomas was significantly load (P < 0.05) than that inadjacent cancers or distal cancers. The specific activities of total AAD in hepatomas was remarkly higher (P < 0. 05) than thatin distal cancers. The melnbrane-combined AADs in hepatoma tissues were increased and no significant difference (P > 0. 05)was found in different tissues of hepatomas, hoar, the soluble molecular form AADs in hepatomaS were significantly higher(P < 0. 05) than that in adjacent cancers or in distal cancers. Conclusion The present data suest that AAD is overexpressed in hepatOIna tissues and is characterized by soluble AAD increment.
