摘 要:目的探讨丁苯肽对血管性痴呆大鼠学习记忆障碍的治疗作用及机制。方法健康雄性Wistar大鼠42只随机分为假手术组、模型组、丁苯肽组,双侧颈总动脉永久结扎法制作动物模型,丁苯肽罐胃,Y-型迷宫评价学习记忆能力,免疫组化检测海马p38MAPK及iNOS表达情况,Tunel染色法检查细胞凋亡。结果丁苯肽组大鼠学习记忆能力强于模型组,低于假手术组,差异均有意义(P<0.05),p38MAPK、iNOS及神经元凋亡较模型组低,高于假手术组,3组两两比较P<0.05。结论丁苯肽能改善VD大鼠的学习记忆障碍,具体可能是通过降低海马p38MAPK、iNOS表达,减少细胞凋亡而起作用。Objective To explore the function and the mechanism of butylphthalide in the treatment of vascular dementia. Methods Forty-two healthy male Wistar rats were randomly alloeated into sham operation group, model group, and butylphthalide group. The models of VD rats were established via occlusion of permanent bi-lateral common carotid arteries. Butylphthalide group was dealt with butylphthalide. Y-type maze test was performed at examining rat's abi[ity of learning and memory. Neuron apoptosis and expression of iNOS and p38MAPK in hippocampus were characterized by TUNEL staining and immunohistochemisty technique,respectively. Results The grades of learning and memory of VD rats were obviously improved by butylphthalide, and the expression of p38MAPK and iNOS was lower than that of model group, but higher than that of sham operation group (P〈0.05). Apoptosis numbers of butylphthalide group were more than those of sham operation group, but less than those of model group. Conclusion Decline in expression of p38MAPK and iNOS and reduction in apoptosis of hippocampus might be involved in butylphthalide's action on improuement of the learning and memory in VD rats.
