摘 要:为建立适于优良品种遗传转化的高效组织培养体系,本研究以3种山东省推广小麦品种(济麦19、良星99、鲁麦14)和模式品种Bobwhite为材料,以成熟胚为外植体对其愈伤组织进行诱导和分化,探讨了2,4-D不同浓度下的3种诱导培养基(MS、N6、MSB5)和4种激素配比的分化培养基对小麦成熟胚离体培养的影响。结果表明,N6培养基的诱导效果最好,平均诱导率超过了80%,且当2,4-D的浓度为4 mg/L时诱导效果最佳。4种激素配比均可诱导小麦成熟胚愈伤组织分化,但分化率较低且处理之间差别较大,添加0.5mg/L IAA和2 mg/L ZT的MS培养基的平均分化率最高,为22.58%。小麦基因型的差别主要影响其分化率的高低而对诱导率的影响不大,3个品种中用于转化的最佳品种为鲁麦14。In order to establish an efficient regeneration system of tissue culture for genetic transformation of improved varieties,the mature embryos of 3 wheat cultivars(Jimai19,Liangxing 99,Lumai 14) planted in Shandong Province and model variety Bobwhite were used as explants to study the callus induction and differentiation;and the effects of 3 induction media(including 3 concentrations of 2,4-D) and 1 differentiation medium(including 4 ratios of hormone)on mature embryos in vitro culture were investigated.The results showed that N6 medium were better than MS and MSB5 for callus induction.Its average induction rate was more than 80%.When the 2,4-D concentration was 4 mg/L,the induction effect was the best.All 4 ratios of hormone could induce the callus differentiation from wheat mature embryos,but the differentiation rate was low and great different.The MS medium supplemented with 0.5 mg/L IAA and 2 mg/L ZT had the highest differentiation rate of 22.58%.The wheat genotype mainly influenced the differentiation rate,while little affected the induction rate.Lumai 14 was the optimum variety for genetic transformation among 3 varieties.
