摘  要:目的探讨RASSF1A表达及其基因启动子甲基化在食管癌发生中的作用。方法取40例食管癌患者肿瘤组织及癌旁组织,用逆转录-聚合酶链反应(RT-PCR)检测RASSF1A的mRNA表达,用甲基化特异性聚合酶链反应(MSP)检测RASSF1A基因启动子的甲基化。结果 RASSF1A mRNA在食管癌组织、癌旁组织表达缺失率分别为77.5%(31/40)、2.5%(1/40),差异有统计学意义(P<0.05)。癌旁组织均未发现甲基化,而癌组织中甲基化率为67.5%(27/40)。所有甲基化的食管癌组织RASSF1A mRNA表达缺失。结论食管癌的发生与RASSF1A基因表达缺失有关联,而RASSF1A基因启动子区甲基化是RASSF1A基因表达缺失的原因。Objective To investigate relationship between RASSF1A expression and promoter methylation of RAS-SF1A gene in esophageal carcinoma.Methods Tumor tissues and adjacent tissues were collected from 40 cases of primary esophageal carcinoma.Expression of RASSF1A mRNA was detected by reverse transcription-polymerase chain reaction(RT-PCR).Methylation of RASSF1A gene promoter was detected by methylation-specific PCR(MSP).Results Expression of RASSF1A mRNA were presented in 39 of normal tissues adjacent to tumor.How-ever,loss of RASSF1A mRNA expression were found in 31 cases in primary tumor.Promoter methylation of RAS-SF1A could be detected in tumor(27 cases) in esophageal carcinoma,while no expression found in normal tissues.In all the esophageal carcinoma cases with promoter gene methylation showed loss expresstion of RASSF1A mRNA.Conclusions There is close relationship between expression of RASSF1A and carcinogenesis of esophageal carcin-oma.Loss expression of RASSF1A gene is due to methylation of the gene promoter.
