慢性粒细胞白血病患者骨髓间质干细胞TGF-β1和SCF表达变化及意义

摘　　要：目的探讨慢性粒细胞白血病(chronic myeloid leukemia,CML)患者骨髓间质干细胞(bone marrowmesenchymal stem cells,BMMSCs)体外干细胞因子(Stem cell factor,SCF)、转化生长因子-β1(transforming growthfactor-β1,TGF-β1)基因表达变化及临床意义。方法 5例慢性期CML患者为实验组,5例健康人为对照组,取骨髓单个核细胞行BMMSCs体外培养,RT-PCR法测定两组BMMSCs的SCF、TGF-β1基因表达量,并分析其相关性。结果慢性期CML患者BMMSCs中SCF基因表达水平较对照组明显升高(P<0.05),而TGF-β1表达水平较对照组明显降低(P<0.01)。实验组内SCF与TGF-β1表达量呈负相关(r=0.948,P<0.05),对照组两指标间无相关性。结论 CML患者BMMSCs的SCF高表达和TGF-β1低表达有相互影响并对白血病细胞的增殖与凋亡发生影响,对CML发病有促进作用。Objective To observe the expression of stem cell factor（SCF）,transforming growth factor-β1（TGF-β1） mRNA of the cultured bone marrow mesenchymal stem cells（BMMSCs） from chronic myeloid leukemia（CML） in myeloid crisis（MC）,and to explore the relationship between the activity change of BMMSCs and pathogenesis of CML.Methods The experimental group included five CML patients with five healthy persons as the control.BMMSCs were cultured in vitro.The expression of SCF,TGF-β1mRNA of BMMSCs were measured by real-time PCR.Results The expression of SCF mRNA in BMMSCs from CML patients was higher（P0.05） and the expression of TGF-β1 mRNA was lower in CML patients than in normal.The expression of SCF had nagtive correlation with the expression of TGF-β1.Conclusion The abnormal express of cytokine such as SCF and TGF-β1 in BMMSCs from CML patients leads to abnormal regulation function to hematopoietic system and affects the cell differentiation and proliferation and apoptosis,which may be closely related to the pathogenesis of CML.
