摘 要:为了防止人们误食河豚鱼而引起食物中毒事件,根据河豚鱼特异性基因,设计并合成引物和相应的TaqMan探针,建立了实时荧光定量PCR检测方法,并应用该方法进行检测试验。结果表明只有河豚鱼和阳性质粒出现扩增曲线,方法的检测灵敏度可以达到100拷贝数左右。该方法具有快速、灵敏和特异等特点,可满足进出口水产品中河豚鱼成分的检测需要。In order to prevent poisonous accident by mistaking poisonous swellfish products, the real-time fluorescence quantitative PCR method was established on the primers and the TaqMan probe designed and synthesized according to the specific DNA sequence of swellfish. The results of real-time fluorescence quantitative PCR showed that only swellfish and the positive plasmids appeared the amplification curve and the detection limitation was about 100 copies of genes. In conclusion, the rapid, sensitive and specific real-time fluorescence quantitative PCR is an effective method for the identification of swellfish materials in aquatic products for qurantine use.
