摘　　要：目的:研究红霉素(ERY)在老年人体内过程的特点.方法:以年轻志愿者为对照,研究ERY在老年人中的药物动力学.10名老年志愿者(老年组)和8名年轻志愿者(年轻组)分别单剂量空腹po ERY肠溶微粒胶囊500 mg后采用微生物法测定血、尿药物浓度,计算药物动力学参数.结果:老年组po ERY 500mg后其c_(max)、t_(max)、T_(1/zβ)和AUC分别为3.04mg/mg/L、2.98h、2.55h和14.58(mg/L)·h,给药后24h时累积尿排出率为4.28%.结论;老年组poERY后吸收较年轻组为缓慢,AUC值较年轻组增高.两组间差异有统计学意义(P<0.05),c_(max)、T_(1/zβ)和尿排出率等两组间差异无统计学意义(P>0.05).AIM:To study the pharmacokinetics of erythromycin in healthy elderly volun-teers. METHODS: The pharmacokinetics of erythromycin in the elderly healthy volunteers were studied and compared with the young healthy volunteers. After oral administration,the drug concentrations in serum and urine were assayed by bioassay,and pharmacokinetics pa-rameters were calculated. RESULTS: After a single oral dose of 500 mg capsules of ery-thromycin, c_(max),t_(max), T_(1/2β) and AUC in the elderly group were 3. 04 mg/L,2. 98 h,2. 55 h and 14. 58 (mg/L) · respectively. The cumulative urinary excretion rate within 24 h was 4. 28%. CONCLUSION: Compared with the young group, the speed of absorption of ery-thromycin was slower,AUC is larger in the elderly group than that in the younger group,the diffe-rences between 2 groups are statistically significant (P<0. 05). However,the differ-ences of other parameters such as c_(max), T_(1/2β) and the cumulative urinary excretion rate between 2 groups are not statistically significant(p>0. 05).
