摘  要:目的探讨吲哚氰绿(ICG)排泄试验与参与肝纤维化的细胞因子-TGFβ1mRNA表达相关性,验证ICG排泄试验可以作为诊断早期肝纤维化的一个敏感指标。方法 42只清洁级Wistar大鼠随机分为正常组(N组)和CCl4组(C组)。C组通过CCl4皮下注射造模,于造模后第2、4、6、8周分别测定ICG15分钟潴留率,用半定量RT-PCR法检测大鼠肝组织TGF-β1mRNA表达。结果成功建立肝纤维化模型,准确测定ICG15分钟潴留率。TGF-β1 mRNA量:正常组、第2、4、6、8周组间TGF-β1mRNA量差异具有显著性(P<0.01);与正常组相比,第4、6、8周组TGF-β1mRNA量差异具有显著性(P<0.01);第2周组TGF-β1mRNA量差异具有显著性(P<0.05),余各组间差异均具有显著性(P<0.01)。结论 ICG排泄试验与肝纤维化的细胞因子-TGF-β1mRNA表达具有良好相关性,该试验可以作为诊断早期肝纤维化的敏感指标。Objective To investigate the indocyanine green(ICG) excretion test with the participation of liver fibrosis in the expression of cytokine-TGFβ1mRNA correlation,ICG excretion test can verify the diagnosis of early stage liver fibrosis as a sensitive indicator.Methods 42 Sprague Wistar rats were randomly divided into normal group(N group) and CCl4 group(C group).C group by subcutaneous injection of CCl4 modeling,the modeling were measured after 2,4,6,8 weeks ICG15 minute retention rate,with semi-quantitative RT-PCR detection of liver tissue expression of TGF-β1mRNA.Results Liver fibrosis model was successfully established,accurate determination of ICG15 minute retention rate.Amount of TGF-β1 mRNA: the control group,2,4,6,8-week group difference between the amount of TGF-β1mRNA significant(P0.01);with the normal group,the first week group TGF-4,6,8 β1mRNA significant amount of difference(P0.01);the first 2 weeks of TGF-β1mRNA significant amount of difference(P0.05),more than differences between groups were significant(P0.01).Conclusion The ICG excretion test of liver cell factor-TGF-β1mRNA expression with good correlation,the test can be used as sensitive diagnostic indicators of early hepatic fibrosis.
