摘  要:目的探讨大黄酸对db/db小鼠的降血糖作用及对胰岛素分泌的影响。方法 30只4周龄db/db小鼠随机分为2组各15只,对照组给予1%纤维素钠,治疗组给予120 mg/kg大黄酸,连续鼻饲灌胃给药8周。投药结束后行经腹腔葡萄糖耐量试验(IPGTT)并测定相应胰岛素水平,其曲线下面积(AUC)代表胰岛素分泌水平,并通过计算IPGTT的0~30 min胰岛素曲线下面积评估早期相胰岛素分泌功能。结果与对照组相比,治疗组糖负荷后0,30,60和120 min的血糖水平明显下降,而30,60和120 min的胰岛素水平明显升高,尤其是早期相胰岛素水平升高更明显。结论早期大黄酸治疗可以明显改善db/db小鼠的葡萄糖耐量,恢复早期相胰岛素分泌功能。大黄酸的这一作用可能使之成为一种新的针对糖尿病的预防或治疗手段。Objective It is to approach the hypoglycemic action of rhein and its' effects on insulin secretion in db/db mice.Methods Thirty 4-week-old db/db mice were randomly divided into two groups with 15 cases in each,the animals were treated with 1% natrium cellulose sodium in control group and with rhein for 120 mg/kg in treatment group.After 8 weeks' treatment,intraperitoneal glucose tolerance test(IPGTT) was performed and AUC(the area under curve) of insulin levels in IPGTT was calculated to evaluate insulin secretory function.AUCINS0-30 was calculated as(INS30-INS0)×15 to evaluate early-phase insulin secretion.Results In treatment group,the blood glucose concentrations at 0 min,30 min,60 min and 120 min after glucose load were significantly reduced,simultaneously measured insulin levels at 30 min,60 min and 120 min were significantly higher than those of control group.The early-phase insulin secretion was remarkably elevated in treatment group.Conclusion Early rhein treatment can significantly improve glucose tolerance,and restore early-phase insulin secretion in db/db mice.The characteristic of rhein may make itself a novel therapeutic means for preventing or curing diabetes.
