摘 要:[目的]了解外源性给予新生雌性BALB/c小鼠己烯雌酚对其脾内淋巴细胞数量和分布的影响以及对T细胞亚型百分比的影响。[方法]选取同一窝、体重差异小于5%的一对雌性新生BALB/c小鼠,随机分为对照组和DES组,共选用25对雌性小鼠,其中DES组新生小鼠在生后24h内颈背部皮下注射DES,每隔24h注射1次,共连续5次,对照组平行注射等量无菌豆油。分别在生后d7、d14、d21、d35和d49将小鼠处死,取脾进行常规石蜡包埋,5μm连续切片,进行CD3和CD20的免疫组织化学检测;用Ficoll分层液分离脾单个核细胞,在流式细胞仪上进行CD4+、CD8+和CD4+CD8+细胞亚型分析。[结果]无论是CD3+细胞还是CD20+细胞,其DES组的平均光密度和积分光密度在生后d7、d14、d21、d35和d49均显著低于对照组。在DES组,CD4+细胞的百分比在生后d35显著降低,在d49显著增高;CD4+CD8+(DP)细胞的百分比在生后d7和d35显著增高,在生后d49显著降低;CD8+细胞的百分比无明显改变。DES组和对照组CD3+和CD20+细胞的脾内分布基本一致。[结论]DES可以显著减少雌性BALB/c小鼠脾内T细胞和B细胞的数量以及T细胞和B细胞表面特异性抗原的表达强度;显著改变CD4+细胞和DP细胞的百分比;但对T细胞和B细胞的脾内分布不产生影响。[Objective]To investigate the effect of diethylstilbestrol administered externally to neonatal female BALB/c mice on the quantity and distribution of their splenic lymphocytes,and on the percentage of T cell subsets.[Methods]Neonatal brood BALB/c mice whose weight discrepancy was less than 5% were divided randomly into control groups and DES groups,and 25 pairs of female mice in all were chosen for use.Mice in DES groups were injected subcutaneously on nape with DES within 24 hours after birth at intervals of 24 hours for 5 times,and mice in control groups were injected with axenic bean oil with the same method.Mice were killed on the d 7,d 14,d 21,d 35 and d 49 after birth separately;the spleens were taken out,imbedded with paraffin and sliced serially in 5 μm.CD3 and CD20 were detected with immunohistochemical technique.Mononuclearcells were separated with Ficoll delamination fluid and CD4+,CD8+ and CD4+CD8+(DP)cells were detected with flow cytometry.[Results]In DES groups the average optical density(AOD)and integrated optical density(IOD)of both CD3+ cells and CD20+ cells on the d 7,d 14,d 21,d 35 and d 49 after birth were all significantly lower than in control groups.In DES groups,the percentage of CD4+ cells was significantly decreased on d 35 after birth and significantly increased on d 49 after birth;the percentage of DP cells was significantly increased on d 7 and d 35 after birth and significantly decreased on d 49 after birth;there were not marked changes were detected on the percentage of CD8+ cells.Distribution of CD3+ and CD20+ cells in spleens of DES groups was similar to that in control groups.[Conclusion]DES can significantly decrease the number of T and B cells and the expressional intensity of specific surface antigen on T and B cells in spleens of female BALB/c mice,significantly change the percentage of CD4+ cells and DP cells,but can not effect the distribution of T and B cells in spleen.
