摘 要:[目的]观察束缚应激对在心肌能量代谢中有重要作用的过氧化体增殖物激活型受体α(PPARα)、肌型肉碱棕榈酰转移酶-I(M-CPT-I)和葡萄糖转运蛋白-4(GLUT-4)mRNA表达的影响。[方法]采用束缚应激模型,按照有无应激和应激时间长短分为正常对照组(C)、束缚1周(R1)、束缚2周(R2)和束缚4周组(R4)。采用逆转录酶多聚酶链反应(RT-PCR)方法检测各组心肌PPARα、M-CPT-I和GLUT-4的mRNA表达水平。[结果]束缚应激1、2、4周大鼠心肌PPARαmRNA和M-CPT-ImRNA水平较正常对照组明显升高(R2组M-CPT-IP﹤0.05,其余P﹤0.01),GLUT-4mRNA水平明显降低(P﹤0.01);束缚应激1、2、4周大鼠各组心肌PPARα、M-CPT-I和GLUT-4mRNA表达无明显差异。[结论]束缚应激上调心肌PPARα和CPT-ImRNA表达,下调GLUT-4mRNA表达。PPA-Rα可能参与束缚应激大鼠心肌能量代谢的改变。[Objective]To explore the effect of restraint stress on the expressions of peroxisome proliferator-activated receptor-α(PPARα),muscle carnitine palmitoyl transferase-I(M-CPT-I)and glucose translator-4(GLUT-4)mRNA in the myocardium of rats.[Methods]Wistar rats were divided into four groups:control,rats under 1 week(R1),2 weeks(R2)or 4 weeks(R4)restraint stress.The expressions of PPARα,M-CPT-I and GLUT-4 mRNA in the myocardium were detected by reverse transcription polymerase chain reaction(RT-PCR).[Results]The expressions of PPARα and M-CPT-I mRNA in the myocardium of rats of the R1,R2 or R4 group were significantly higher than that of control group(R2 group M-CPT-I P﹤0.05,others P﹤0.01),while the expression of GLUT-4 mRNA in the myocardium of rats of the R1、R2 or R4 group was significantly lower than that of control group(P﹤0.01).There were no significant differences in peroxisome proliferator-activated receptor-α(PPARα),muscle carnitine palmitoyl transferase-I(M-CPT-I)and glucose translator-4(GLUT-4)mRNA expressions of the myocardium between R1,R2 or R4 restraint stress group rats.[Conclusion]Restraint stress up-regulates the expressions of PPARα,M-CPT-I and down-regulates the expression of GLUT-4 mRNA in the myocardium of rats.PPARα may involve in the change of energy metabolism in the heart under restrain stress.
