摘 要:目的研究食管癌患者外周血中TACSTD1和CK19的表达,探讨其作为食管癌微转移标志物的可行性和其对食管癌患者预后的影响。方法用实时荧光定量RT-PCR联合检测30例食管癌患者手术切除的肿瘤标本和食管残端TACSTD1和CK19的表达并比较二者的区别,检测30例健康献血员外周血标本TACSTD1和CK19的表达得出正常人群的表达范围,以此为标准检测30例食管癌患者术前外周血标本是否存在TACSTD1和/或CK19的高表达。结果 TAC-STD1和CK19在食管癌组织中的表达量均明显高于食管残端组织(P<0.01)。22例患者的外周血标本检测出TACSTD1和/或CK19的高表达。CK19和TACSTD1在外周血中的表达量与患者的肿瘤分化程度高低(P<0.05)和T分期(P<0.05)、N(P<0.01)分期有密切联系。结论 CK19和TACSTD1在食管癌组织均有特异性的高表达,在食管癌的发生、发展中可能起着重要的作用。CK19和TACSTD1是有价值的食管癌微转移标志物。Objective To study the expression of CK19 and TACSTD1 in peripheral blood of patients with esophageal cancer, and explore the possibility to use it as a marker for micro-metastasis of esophageal cancer and its influence on prognosis of esophageal cancer patients. Methods Expression of CK19 and TACSTD1 was detected in 30 tissue samples from esophageal cancer patients and in 30 peripheral blood samples from healthy volunteers by real-time quantitative reverse transcription polymerase chain reaction (QRT-PCR). Results The expression level of CK19 and/or TACSTD1 was significantly higher in esophageal cancer tissue samples than in its stump tissue samples(P0.01). The TACSTD1 and /or CK19 were highly expressed in peripheral blood samples from 22 patients. The expression level of TACSTD1 and CK19 was closely related with the severity of tumor differentiation(P0.05), T (P0.05) and N stages(P0.01). Conclusion CK19 and TACSTD1 are highly expressed in esophageal cancer tissue and play an important role in occurrence and development of esophageal cancer. CK19 and TACSTD1 are good markers for micro-metastasis of esophageal cancer.
