摘  要:目的研究石蒜碱对裂殖酵母生长周期的影响,分析石蒜碱的抗肿瘤机制,推测用裂殖酵母作为模型筛选抗肿瘤药物的可行性。方法在不同浓度石蒜碱条件下培养裂殖酵母,测量生长曲线;流式细胞术检测细胞周期分布;光学显微镜观察形态,DAPI及Calcofluor染色观察细胞核、细胞间隔变化;Real-time PCR检测细胞周期相关基因的表达。结果在试验条件下,石蒜碱显著抑制裂殖酵母生长,并呈现时间浓度依赖性,作用20.5 h后IC50值为212μmol/L;酵母细胞变长、细胞核及细胞间隔增多,G2/M期细胞比例增加,部分细胞周期相关基因在mRNA水平有所变化。结论石蒜碱对裂殖酵母生长具有明显抑制作用,可引起细胞表型变化,影响部分细胞周期相关基因的表达。Objective To study the impact of Lycorine on the growth of Saccharomyces Pombe,to speculate the anticancer mechanism of Lycorine,and then to discuss the practicability of screening anticancer drugs by fission yeast.Methods S.Pombe strains were cultured under different concentrations of Lycorine in order to measure its growth curve.The cell-cycle distribution was detected by flow cytometry,and the cellular morphology was observed by microscope after DAPI and Calcofluor fluorescent staining.The expression levels of some cell-cycle related genes were detected by Real-time PCR.Results Under the experimental condition,the growth of S.Pombe was remarkably inhibited by Lycorine in time-dependence and concentration-dependence.The proportion of G2/M cells largely increased to 212 μmol/L after 20.5 h,and the expression levels of some important cell-cycle related genes were changed.Conclusions Lycorine can inhibite the growth of S.Pombe,change the cellular morphology,and affect the expressions of some cell-cycle related genes.
