燕麦细胞癌裸小鼠移植瘤中C型病毒的电镜诊断

摘　　要：一株人肺燕麦细胞癌成功地移植到ＢＡＬＢ／ＣＡ／ＪＣＬ品系裸小鼠皮下生长并连续传代。电镜跟踪观察显示，传代早期移植瘤较好地保持了来源瘤的超微结构基本特征，最典型的结构是在移植瘤细胞胞浆中出现以成堆分布为主的神经分泌型颗粒。但是在第十八代移植瘤的细胞间隙以及胞浆中小胞和内质网小池内查见到Ｃ型病毒颗粒。神经分泌型颗粒与成熟的Ｃ型病毒在电镜下形态相似，即都有外膜、透明带和致密内核，粒径在１００ｎｍ左右。但两者还是有细微差别：首先是粒径范围不尽一致。其次是分布区域有所不同。此外，裸小鼠内源性Ｃ型病毒活化、侵染人癌移植瘤尚有其它特异性形态表现。本文还探讨了人肺燕麦细胞癌激活Ｃ型病毒的可能机理。A human lung oat cell carcinoma was transplanted and serially passaged in BALB/CA/JCL nude mice, cultivated under a specific pathogen free condition. The ultrastructural characteristics of original tumor were maintained in passages from 1 to 14. The most striking feature was the neurosecretory type granules (NSG) which were often presented in the cytoplasmic processes and beneath the cell membrane. In the 18th generation, the C type viruses were found in extracellular space and within the cytoplasmic vacuoles and the cisternae of endoplasmic reticulum. The electron microscopic image of NSG was similar to that of mature C type virus:both of them with a similar outer diameter of about 100nm, which were composed by the limiting membranes, a transparent and a dense cors. But the diameter range and distributional zone of C type viruses were different from NSG. Sometimes the typical features of C type viruses may been found when they were activated or were invading the human hetereotransplanted tumor. In addition, the activated mechanism of endogenous C type of nude mice was discussed.
