摘  要:目的探讨小鼠体内雄激素水平缺乏对心脏衰老的影响。方法实验分为正常C57BL/6 J雄鼠组(n=8)、去势组(n=8)、睾丸女性化鼠(tfm)组(n=10)、衰老组(n=8)。测定各组血清睾酮浓度,分离心肌组织测定组织超氧化物歧化酶(SOD)和丙二醛(MDA)水平以及W estern印迹测定去磷酸化Rb蛋白表达量。结果 与正常组相比,衰老组、去势组和tfm组血清睾酮浓度明显下降(P<0.01);心肌组织中SOD活性下降(P<0.05,P<0.01)、MDA含量增加(P<0.01);去磷酸化Rb蛋白表达量增加(P<0.01)。与衰老组相比,去势组和tfm组SOD活性、MDA含量及去磷酸化Rb蛋白表达量差异无统计学意义。结论体内雄激素水平缺乏可能通过增加活性氧水平(ROS)及Rb蛋白表达导致小鼠心肌组织衰老。Objective To explore the effect of androgen deficiency on myocardium senescence in mice.Methods At 8 weeks of age,male C57BL/6 mice were randomly separated into control,castrated and senescence groups,each group had 8 mice.Tfm mice were used as tfm group(n=10).Besides senescence group,all groups were fed for 24 w.Blood was collected for measurement of testosterone concentrations.The activity of SOD and the level of MDA in the myocardium tissues were determined,and the expression of dephosphorylation Rb was detected by Western blot.Results Compared with those of control group,the lower testosterone concentrations(P〈0.01),the lower SOD activity(P〈0.05) and higher MDA level(P〈0.01),dephosphorylation Rb expression(P〈0.01)were observed in the senescence, castrated and tfm groups.Compared with senescence group,these changes had no significant difference between castrated and tfm groups.Conclusions Testosterone deficiency is associated with myocardium senescence and this change may result from the increased ROS level and Rb protein expression in mice.
