摘  要:研究一种新型双光子荧光素(DMAHAS)对人肝癌HepG2细胞的毒性及肿瘤细胞标记后的体内示踪。体外细胞毒性分析采用MTT、中性红(NR)、考马斯亮蓝(CB)和流式细胞术(FCM)等方法。DMAHAS标记肿瘤细胞的体内示踪在人肝癌模型中进行,切除的肿瘤异种移植物经荧光成像和传统的组织病理分析。此外,我们建立了一种基于DMAHAS释放的细胞毒性分析方法。研究结果表明DMAHAS对HepG2细胞无明显毒性,它显示出对活细胞很高的穿透性和稳定的细胞质定位。体内细胞示踪实验表明它是一种用于肿瘤示踪和荧光成像的可靠探针。To explore the cytotoxicity of a two-photon absorbing fluorescence(DMAHAS) on human HepG2 cells and its application for tracking the tumor cell in mice bearing human HepG2 cells.The in vitro cytotoxicity of DMAHAS were evaluated by using MTT,neutral red(NR) assay,coomassie brilliant blue(CB) assay and flow cytometric analysis(FCM).The application of DMAHAS for tumor tracking in vivo was conducted in a human liver tumor xenograft mice model.Tumor xenografts were examined by fluorescence imaging and conventional histological methods.The results showed that DMAHAS had no significant cytotoxicity on HepG2 cells.It showed high cell-permeability to live cells with the stable cytosolic localization.In vivo study suggested that it was a reliable probe for tumor tracking and fluorescence imaging.
