摘 要:微生物几丁质酶在生物防治中有着重要的应用价值,发酵条件的优化是大量获取几丁质酶的重要前提;试验利用单因素优化法研究了影响地衣芽孢杆菌JDZ-3产生几丁质酶的培养基的初始pH、碳源、氮源、培养温度、金属离子,利用正交试验确定了发酵培养基的组成;培养基的最佳初始pH为7.0,几丁质酶的最佳碳源为胶体几丁质,最佳氮源为酵母膏,最佳金属离子为Mg2+,最适的发酵温度为30℃,该菌株产几丁质酶发酵培养基的三大营养元素的组成为:胶体几丁质0.2%,酵母膏0.6%,MgSO4·H2O0.1%;在此优化条件下,地衣芽孢杆菌JDZ-3产几丁质酶的能力明显提高,其发酵上清液中的几丁质酶的活力可达2.76U/mL。Microbial chitinase has important value in biological control. The optimum fermentation condition is a prerequisite for increasing of chitinase production. The compositions of the culture medium for Bacillus licheniformis JDZ-3 to produce chitinase were studied by single factor experiments. The compositions of the culture medium included the best medium of initial pH, carbon source, nitrogen source, culture temperature and metal ions. Three major nutrients made up the chitinase fermentation medium were studied by the orthogonal experiments. The main factors which determined chitinase production as follows: the optimum initial medium pH was 7.0, the best carbon source in the medium was the colloidal chitin, the best nitrogen source was yeast extract, the best metal ion was Mg 2 + , and the optimal fermentation temperature was 30℃ . In the orthogonal experiments based on the single factor optimization method, three major nutrients made up the chitinase fermentation medium were obtained: 0.2% colloidal chitin, yeast extract 0.6%, MgSO 4 ·H 2 O 0.1 %. The chitinase production capacity by Bacillus licheniformis JDZ-3 significantly increased in optimum conditions. The strain cultured with this fermentation medium, the chitinase in the supernatant was up to 2.76 U/mL.
