摘 要:为探讨猪链球菌能否成为单增李斯特菌四环素耐药基因tetM的水平传播宿主,以携带该基因的单增李斯特菌四环素耐药菌株为供体菌株,猪链球菌的红霉素耐药菌株为受体菌株进行滤膜接合试验。同时对供体菌株进行了转移相关基因int-Tn的PCR检测。结果显示,获得19个接合子,接合转移率为4×10-7。接合子均对四环素耐药,且接合子中均PCR扩增到tetM基因,该基因克隆测序结果与供体菌中tetM基因序列完全一致。同时,从供体菌株中扩增到整合酶编码基因int-Tn。这说明猪链球菌可以成为单增李斯特菌四环素耐药基因tetM的水平传播宿主,并且该基因的水平转移与整合子有密切关系。To explore the possibility of Streptococcus suis as the horizontal transmission host of tetracycline resistant gene tetM in Listeria monocytogenes,the filtermating experiment was performed in which tetracycline-resistant L.monocytogenes strain carrying tetM gene was used as the donor strain and erythromycin-resistant S.suis strain as recipient strain.The int-Tn gene related to transfer of tetM gene was detected from the donor strain by PCR.19 transconjugants resistant to tetracycline were obtained at a frequency of 4×10-7.The tetM gene was amplified from transconjugants,which shared 100% homology in nucleotides with tetM gene from donor strain.The int-Tn gene that encoded integrase was amplified from the donor strain.The results demonstrated that S.suis could become the horizontal transmission host of tetracycline resistant gene tetM in L.monocytogene,and horizontal transfer of tetM gene was closely related to integron.
