摘 要:为了解云南省昆明市屠宰猪戊型肝炎病毒(HEV)感染情况,对68份从农贸市场和屠宰场采集的猪肝样品,应用免疫组织化学染色、RT-PCR分别对HEV的抗原和核酸进行了检测,并对肝样品进行了组织病理学观察。免疫组织化学染色结果表明,83.8%(57/68)的肝呈HEV抗原阳性。组织病理学观察发现,47.1%的肝组织出现炎性细胞浸润,22.1%的肝出现肝细胞变性、坏死,20.6%出现肝细胞萎缩。Mal-lory三色染色法染色结果显示,有25%的猪肝存在不同程度的结缔组织增生。RT-PCR结果显示,有1份肝呈HEV核酸阳性,阳性率为1.5%。进化分析表明,这株病毒属4b基因亚型,与广西猪源、越南人源毒株的同源性最高,证实HEV的流行具有地理分布的特征。Immunohistochemical staining showed that 68 swine liver samples,which were collected from slaughter house and farm produce market in Kunming,Yunnan Province,had sporadically hepatitis E virus(HEV) antigen in hepatocyte,and the HEV positive rate was 83.8%(57/68).Histopathologic observation showed that sporadic denaturation and necrobiosis(22.1%) and karyopynosis(20.6%) of hepatocyte and infiltration of inflammatory cells(47.1%) occurred in the liver samples.Mallory staining showed that fibrous connective tissue proliferation was observed in 17 of the 68 samples.HEV RNA was detected in 1 of the 68 samples(1.5%) by RT-PCR.Phylogenetic analysis showed that the HEV isolated from one of the 68 samples was classified into subgenotype 4b and shared highly homologous with the swine origin HEV isolates prevalent in Guangxi Province of China and the human origin HEV strains HE-JVN1-FS and HE-JVN1 prevalent in Vietnam,indicating that HEV prevalence was characterized by geographical distribution.
