经鞘内注射pLXSN—bcl-2对缺血再灌注大鼠脑梗死体积及caspase-3蛋白的影响

摘　　要：目的：研究通过鞘内注射质粒pLXSN-bcl-2在脑缺血再灌注大鼠中的脑保护作用. 方法：60只Wistar大鼠随机分为生理盐水组、pLXSN组和pLXSN-bcl-2组,以改良的线栓法制作大脑中动脉闭塞2h模型,经鞘内注射生理盐水、pLXSN、和pLXSN-bcl-2,在I/R后24h、72h处死大鼠,检测脑梗死体积、bcl-2、caspase-3蛋白表达.结果：MCAO 24h、72h pLXSN-bcl-2组脑梗死体积及caspase-3蛋白与生理盐水组、pLXSN组比较减少,P〈0.05, pLXSN-bcl-2组bcl-2蛋白表达与生理盐水组、pLXSN组比较阳性细胞百分比显著增高P〈0.01.结论：通过鞘内注射质粒pLXSN介导bcl-2基因在I/R大鼠中抑制caspase-3蛋白表达、减少脑梗死体积,起到脑保护作用,鞘内注射是安全有效的途径.Objective clarify rat brain protection of intrathecal injection pLXSN-- bcI--2 gene in cerebral ischemia--reperfusion. Methods,60 Wistar rats were randomly divided into three groups, normal saline group, pLXSN group and pLXSN--bcl--2 groups, with improved suture method of middle cerebral artery occlusion 2h model, were treated with intrathecal injection of saline, pLXSN, or pLXSN--bcl--2. The I / R rats were killed after 24h or 72 h, and detected cerebral infarct volume, bcl-- 2, caspase-- 3 protein expression. Results ： infarction volume , caspase-- 3 expression of pLXSN-- bel-- 2 group are less than saline and empty plasmicl group after 24h and 72h, P d0.05, the percentage of bcl-- 2 positive cells in pLXSN-- bcl-- 2 group was remarkable increased compare to empty plasmid and normal saline group, P d0.01. Conclusion; Intrathecal injection of pLXSN- bcl-2 gene could effective protect I/R brain, by inhibiting caspase-3 protein expression, decreasing infarct volume. Intrathecal injection is effective procedure.
