摘 要:目的:在体外环境下观察基因重组人生长激素(rhGH)及联合氟尿嘧啶(5-FU)干预不同细胞膜生长激素受体(GHR)表达水平对人结肠癌细胞的增殖情况。方法:采用免疫细胞化学法检测人结肠癌细胞株HCT-8和LOVO膜表面的GHR表达水平。每株细胞分为对照组、rhGH组、5-FU组和rhGH+5-FU组,用四甲基偶氮唑蓝(MTT)、克隆形成实验和免疫细胞化学等方法,测定各组各株肿瘤细胞的抑制率、增殖细胞核抗原(PCNA)表达变化。结果:HCT-8高表达GHR(阳性表达细胞数≥50%),LOVO未表达GHR。rhGH显著提高HCT-8细胞增殖和克隆形成率,PCNA表达增加,且联合化疗后,较LOVO组肿瘤抑制率减少(P<0.05)。结论:在体外环境下,rhGH促GHR阳性表达的结肠癌细胞增殖,且对5-FU化疗有抵抗,对于不表达GHR的结肠癌细胞无上述作用。Objective: To investigate the effect of recombinant human growth hormone(rhGH) and 5-FU on the growth of colon cancer cells with dfferent expression of growth factor receptor(GHR) in vitro.Methods: The expression level of membrane GHR of colon cancer cell line HCT-8 and LOVO were measured by immune cellular chemistry.The two cell lines were divided into 4 different groups according to the different treatment as indicated:control group,rhGH group,rhGH+5-FU group and 5-FU group.Cell growth/inhibition rates were determined by MTT,colony formation and PCNA were measured by immune cellular chemistry.Results: HCT-8 showed high level GHR(≥50%),LOVO showed no GHR expression,and rhGH increased the growth ability of HCT-8 cells.Compared with LOVO,HCT-8 cells showed less growth inhibition under the stimulation of rhGH and more PCNA expression.Conclusion: rhGH can stimulate GHR positive colon cancer cell growth,protect the cell growth inhibition from chemoraphy,and this effect was not observed in GHR negative colon cancer cells.
