摘 要:目的探讨重组人生长激素(rhGH)及联合5-氟尿嘧啶(5-FU)对人结肠癌荷瘤裸小鼠的影响。方法建立人结肠癌LOVO裸小鼠皮下移植瘤模型,将荷瘤裸小鼠随机分为对照组、GH组、5-FU组、GH/5-FU组,每组8只,同时治疗10天,观察裸鼠体重、皮下移植瘤体积、重量的变化,以流式细胞术测定细胞周期及凋亡情况。将另外32只小鼠编为第二大组,同法分为4组,并分别给于上述处理,观察生存期。结果与对照组比较,GH组裸鼠的体重明显增加(P<0.05);与5-FU组比较,GH/5-FU组裸鼠的体重明显增加(P<0.05)。与对照组比较,5-FU组、GH/5-FU组肿瘤体积、重量明显减少(P<0.05)。与5-FU组比较,GH/5-FU组裸鼠的肿瘤体积、重量差异无统计学意义(P>0.05)。与对照组比较,GH组S、G2/M期细胞百分比及增殖指数(PI)差异无统计学意义(P>0.05)。与5-FU组比较,GH+5-FU组的S、G2/M期细胞百分比及PI差异无统计学意义(P>0.05)。与对照组比较,5-FU组、GH+5-FU组的凋亡率均升高(P<0.05)。与5-FU组比较,GH+5-FU组的凋亡率差异也无统计学意义(P>0.05)。与对照组比较,5-FU、GH/5-FU组明显延长(P<0.05);与5-FU组比较,GH/5-FU组延长,但无统计学意义(P>0.05)。结论生长激素在荷瘤裸鼠体内不刺激肿瘤生长,对机体也无不良影响。联合化疗时,未发现增敏现象。Objective To investigate the effects of recombinant human growth hormone(rhGH) with 5-fluorouracil(5-FU) on human colon carcinoma.Methods Human colon carcinoma LOVO model was established in nude mice.Thirty-two nude mice were divided into 4 groups: control group,GH group,5-FU group,GH/5-FU group.Ten days after treatment,the body weight,tumor weight and volume of each group were observed.The cell cycle,proliferation index(PI) and rate of apoptosis were analyzed by flow cytometry.The survival time was observed on other 32 nude mice which were grouped and treated in the same way.Results Compared with the control group,the body weight of GH group was significantly higher(P0.05);the body weight of GH/5-FU group was higher than that of 5-FU group(P0.05).Compared with the control group,the tumor mass and volume were significantly reduced in 5-FU group and GH/5-FU group(P0.05),there were no significant differences between GH/5-FU group and 5-FU group(P0.05).The percentages of S and G2/M phase,and proliferation indices(PI) were not significantly changed in GH group compared with control group(P0.05),and there were no differences between GH/5-FU group and 5-FU group(P0.05).The rate of apoptosis in 5-FU and GH+5-FU groups was increased significantly compared with that of control group(P0.05),and there were no significant differences between GH/5-FU group and 5-FU group(P0.05).The survival time of 5-FU and GH+5-FU groups was increased significantly compared with control group(P0.05) and there were no differences between GH/5-FU group and with 5-FU group(P0.05).Conclusion rhGH may not stimulate tumor growth and not have adverse effects;it may not affect the sensitivity of chemotherapy significantly in tumor-bearing nude mice.
