摘  要:综述了近年来有关癌症中自吞噬作用的相关文献,对通过控制自吞噬过程来作为一种新的癌症治疗手段的意义和可行性进行了回顾,认为自吞噬是细胞内长寿蛋白和细胞器进行降解/再循环的主要通路,是一种真核细胞中广泛存在、进化上高度保守的过程,参与细胞的生长和内环境稳态等的调节.多种细胞内外的环境刺激能够启动自吞噬机制,如饥饿、激素、药物、细菌感染、蛋白质的聚集或错误折叠和细胞器的损伤等.自吞噬作为另外一种有别于凋亡的细胞程序性死亡概念的提出使其在肿瘤发生、发展中的作用得到高度关注,成为肿瘤研究领域的一个热点.In this paper, the significance and feasibility of modulating autophagy as a new and promising treatment for cancer therapy are reviewed, based on recent research reports of autophagy associated with cancer. Autophagy is a major intracellular degradation/recycling pathway for long - lived proteins and organelles. It takes place in all eukaryotic cells and is a highly conserved and well - regulated process, playing a central role in the regulation of cell growth and homeostasis. Variety of intracellular and extracellular stress conditions can trigger dramatic enhancement of autophagy, such as starvation, hormones, drugs, infection, aggregative or misfolding proteins, damaged organelles, etc. In recent years, a growing number of studies have revealed that autophagy plays a critical role in tumor generation, progress and metastasis as a distinguished programmed cell death from apoptosis. Therefore, as a promising strategy, auto phagy has attracted considerable attention from researchers in tumor therapy.
