摘  要:目的:研究黄精Polygonatum sibiricum不同分子量级的多糖组分对α-葡萄糖苷酶的抑制作用,探究黄精多糖分子量大小、单糖组成、单糖比例和黄精对α-葡萄糖苷酶抑制作用之间的联系。方法:采用DEAE2纤维素柱色谱和透析袋透析等操作对黄精的醇提物进行分离纯化分段。然后用气相色谱分析多糖不同分子量级组分的组成、单糖比例。结果:黄精中抑制α-葡萄糖苷酶活性的主要成份为黄精多糖。对黄精多糖分离纯化分段,得到4个分子量级别的多糖,对其组成分析:M<2 000,含有2种单糖,分别为阿拉伯糖、葡萄糖,对α-葡萄糖苷酶的抑制率为44.90%;2 000<M<14 000,含有一种单糖为葡萄糖,对α-葡萄糖苷酶的抑制率为68.10%;14 000<M<50 000,含有3种单糖,分别为甘露糖、葡萄糖、半乳糖,对α-葡萄糖苷酶的抑制率为55.30%;M>50 000,含有2种单糖,分别为甘露糖、葡萄糖,对α-葡萄糖苷酶的抑制率为63.80%。结论:单一葡萄糖形成的黄精多糖抑制α-葡萄糖苷酶活性最强,降糖活性也最强。AIM: To study the inhibitory activity of different molecular-level polysaccharide from Polygonatum sibiricum on α-glucosidase and explore the relationship among their molecular weight monosaccharide composition effect of monosaccharide on α-glucosidase.METHODS: DEAE2 cellulose column chromatography and dialysis bags of dialysis operations were used to isolate and purify the alcoholic extract.Then GC was assigned to analyze the composition,proportion of monosaccharide of different molecular-level.RESULTS: Solomonseal polysaccharide of inhibitory activity on α-glucosidase was composed of four molecular-level polysaccharide,in cluding MW 〈 2 000,in the form of arabinose and glucose,the inhibition rate was 44.90%;2 000 〈 MW 〈 14 000,in the form of glucose,the inhibition rate was 68.10%;14 000 〈 MW 〈 50 000,in the form of mannose,glucose and galactose,the inhibition rate was 55.30%;MW 〉 50 000,in the form of mannose and glucose,the inhibition rate was 63.80%.CONCLUSION: Solomonseal polysaccharide which is formed by single glucose has the strongest inhibitory activity on α-glucosidase,meanwhile,polysaccharide has the strongest hypoglycemic activity.
