摘  要:目的研究降糖孜亚比提片(Hypoglycemic Ziyabiti Tablet,HZT)对2型糖尿病(T2DM)大鼠血糖和血脂的影响。方法 (1)考察HZT的急性毒性。(2)采用高脂乳剂给Wistar大鼠灌胃10d后,用毛细管法判定胰岛素抵抗性;再结合两次给药法腹腔注射四氧嘧啶(alloxan,ALX),建立类似于人类T2DM的大鼠模型。将大鼠分为6组:正常组、模型组、消渴丸阳性组(420mg/kg)(XKP组)及HZT低、中、高剂量组(220、440、880mg/kg)。(3)观察HZT对T2DM模型大鼠空腹血糖(FBG)、血清胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白(HDL-C)、空腹血清胰岛素(FINS)、胰岛素分泌指数(IS)、胰岛素敏感指数(ISI)等生化指标的影响。结果 (1)在给予小白鼠相当于人体临床用量540倍剂量时,未出现急性毒性。(2)灌胃高脂乳剂后,大鼠KITT值由正常(5.8±0.5)下降为(2.8±0.9)(P<0.01)。(3)与模型组比较,HZT低、中剂量组FBG和TC明显降低,差异具有统计学意义(P<0.05);HTZ中剂量组和消渴丸阳性组(XKP)空腹血清胰岛素(FINS)水平升高,差异具有统计学意义(P<0.01);HZT低、中剂量组和XKP组IS均升高(P<0.05)。结论 HZT能降低T2DM模型大鼠FBG和TC,具有促进细胞消耗葡萄糖的作用,且安全有效。提示其降血糖机制可能是通过促进胰岛素分泌、增加细胞对葡萄糖的利用而起作用的。Objectives To study the effects of Hypoglycemic Ziyabiti tablet(HZT)extracts on type 2 diabetes mellitus rats and observe its exoteric activity.Methods The acute toxicity of HZT on mice was observe.Wistar rats were administrated with high fat emulsion for 10 days,assessed the insulin resistance by using blood capillary method.They were injected alloxan(ALX)to abdominal cavity in continuous two days,and then the type 2 diabetic rat model was established.The rats were divided into 6 groups:normal group,model group,positive group of Xiaoke Pills(420 mg/kg)(XKP)and low,middle,high dose groups of HZT(220,440,880 mg/kg).The effects of HZT on T2DM rats fasting blood glucose(FBG),total cholestero(TC),triglyeride(TG),high densitv lipoprotein(HDL-C),FINS,insulin secretion(IS),insulin sensitivity index(ISI)were observed.Results(1)At a dosage of 540 times greater than used in humans,there were no acute toxicity.(2)After administrated with high fat emulsion,the KITT of Wistar rats decreased from 5.8±0.5 in normal control to 2.8±0.9(P0.01).(3)Compared with model group,low and middle dose groups of HZT decreased the T2DM rats' FBG and TC(P0.05);middle dose group of HTZ and positive group of Xiaoke Pills(XKP)increased FINS(P0.01);low and middle dose groups of HZT and XKP group enlarged IS(P0.05).Conclusion HZT could decrease the T2DM rats' FBG,TC and had the effective of promoting cell consuming glucose,which hinted the hypoglycemic mechanism was possibly to promote insulin secretion and increase the utilization of glucose.
