摘 要:目的探讨转化生长因子β(TGF-β)通路中的转化生长因子βⅠ型受体(TβRⅠ)和Smad4蛋白在肝内胆管细胞癌组织中的表达及其可能的作用机制。方法应用免疫组织化学EnVision二步法,检测19例正常肝组织肝内胆管细胞,33例肝内胆管细胞癌组织中TβRⅠ、Smad4蛋白的表达情况。结果 33例肝内胆管细胞癌组织中TβRⅠ和Smad4蛋白的阳性表达率分别为30.30%、24.24%,19例正常肝组织肝内胆管细胞中TβRⅠ和Smad4蛋白的阳性表达率分别为68.42%、63.16%,肝内胆管细胞癌组织中TβRⅠ和Smad4表达明显低于正常肝组织肝内胆管细胞,差异有统计学意义(P<0.01)。肝内胆管细胞癌组织中TβRⅠ蛋白表达与Smad4蛋白表达呈正相关(r=0.232 4,P<0.01)。结论 TβRⅠ和Smad4蛋白在肝内胆管细胞癌组织中的表达与肝内胆管细胞癌的发生、发展密切相关。Objective To explore the expressions of TβR Ⅰ and Smad4 in TGF-βpathway andpossible mechanisms by which they affect primary intrahepaticcholangiocarcinoma.Methods EnVisionimmunohistochemistry was used to detect the expressions of TβR Ⅰ and Smad4 protein in 33 patients withPIC and 19 normal intrahepatic bile duct tissues.Result The positive rates of TβR Ⅰ and Smad4 proteinwere 30.30%,24.24% in 33 patients with PIC respectively;The positive rates of TβR Ⅰ and Smad4protein were 68.42%,63.16% in 19 normal Intrahepatic bile duct tissues,respectively;the expressionsof TβR Ⅰ and Smad4 in primary intrahepaticcholangiocarcinoma were significantly lower than in normalIntrahepatic bile duct tissues(P〈0.01).The correlation analysis indicated that the expression of Smad4in primary intrahepaticcholangiocarcinoma was positively related to that of TβR Ⅰ.Conclusion Theexpressions of TβR Ⅰ,Smad4 protein were closely related with the occurrence and progression of primaryintrahepaticcholangiocarcinoma.
