摘 要:目的:应用倒置荧光显微镜动态观察变形链球菌(变链菌)Ingbritt C国际标准株及luxS基因缺陷株不同时期生物膜形成的差异。方法:建立以玻片为载体的生物膜模型,分别取两菌株6、18、24、48h的生物膜,观察不同时期两菌株生物膜与玻片的黏附程度,吖啶橙(AO)荧光染色后,倒置荧光显微镜下及时观察生物膜形态及细菌之间相对活动度,并对不同时期的生物膜标本进行处理和保存,观察3个月后倒置荧光显微镜下生物膜形态有无改变。结果:应用倒置荧光显微镜可对孔板内生物膜进行直接动态观察,镜下两菌株生物膜形态多样;6h开始出现细菌聚集成膜,18~24h形成最佳,与玻片黏附牢固而不易冲掉;48h标准株生物膜附着于玻片能力较缺陷株弱,易整体滑落;两菌株生物膜标本经处理保存3个月后镜下形态未发生改变。结论:变链菌标准株与luxS基因缺陷株生物膜具有不同的空间立体结构,形态多样,且在不同时期、生物膜形态、细菌间的相互作用及黏附力不同。Objective:To study the difference of biofilm formation at different times by dynamic observation of converted fluorescence microscope between Streptococcus mutans Ingbritt C international standard strain and LuxS mutant strain.Methods:The biofilm models of both Streptococcus mutans standards strain and luxS mutant strain were built on the slide carriers in 6,18,24,48 h respectively,in order to study the adhering levels between the biofilm and slides of two kinds of strains in different periods.Then the biofilm specimens were processed and preserved for three months,observing whether there was any change in the microscopic appearances of the specimens.The biofilms of the two strains and the relative mobility were observed using converted fluorescence microscope after dyeing by means of acridine orange(AO) between the bacteria.Results:The biofilms in the microtiter plates could be observed dynamically by converted fluorescence microscope.There were varieties in morphology of the two stains.The biofilms began to form the bacteria gathering densely at 6 h.Those of 18 h and 24 h were the best formation.They can adhere to slide carrier very firmly and can not be washed out.At the time of 48 h,the adhesive capacity of the standard strain was lower than that of luxS mutant strain.The whole biofilm of the standard strain was prone to slide.There were no changes in biofilm specimens of two kinds of strains in different periods after three months.Conclusion:The biofilms of Streptococcus mutans standard strain and luxS mutant strain were three-dimensioned and variety in morphology.In addition,at different point-in-times,they displayed a marked difference in the form of biofilm,reciprocity of bacteria and adhesive power.
