摘　　要：目的:探讨油菜花粉脂溶性提取物(BPE)对体外培养的人肺腺癌细胞A549的杀伤效应。方法:以人肺腺癌细胞A549为研究对象,电镜观察用药前后A549细胞形态,应用CCK-8法检测BPE对A549细胞增殖的影响,用流式细胞术检测BPE对A549细胞周期和凋亡的影响。RT-PCR检测bax/bcl-2基因表达。结果:BPE对A549细胞增殖有抑制作用,在一定浓度范围内,呈量-效关系,当BPE浓度为1000μg/mL时,A549细胞增殖的抑制率达到最高,为76.54%。透射电镜下可见细胞核固缩,染色质凝集或边聚,胞浆内出现空泡,并有凋亡小体出现。流式细胞仪检测结果显示A549细胞周期比例分布出现变化,G0/G1期细胞比例呈下降趋势,而S期和G2-M期比例则上升;随着时间的延长,细胞凋亡率上升。RT-PCR检测显示BPE的作用效应不是通过bax/bcl-2基因实现的。结论:BPE可抑制人肺腺癌细胞A549的增殖,诱导细胞的凋亡。Objective To observe the effects of brassica campestris pollen extract（BPE） on human lung adenocarcinoma cell line A549 in vitro. Methods Human lung adenocarcinama cell line A549 was used as study object,observed the cells morphology of A549 by electron microscopy,the cytotoxicities of BPE on A549 cell was evaluated by CCK-8 assay. To observed the effect of BPE on cell cycle and apoptsis of the A549 by flow cytometry. Expression of bax/bcl-2 gene was detected by PR-PCR. Results In a certain concentration range,BPE could inhibit A549 cell proliferation in a concentration-dependent pattern. The killing effect was the biggest when the dose of BPE was 1 000 μg/mL,the inhibit rate was 76.54%. Changes of A549 cell＇s ultrastructure was observed by electron microscopy：cellular shrinkage,nuclear condensation,chromatin margination,membrance blebbing,and apoptotic boby appearing. FCM assays showed that the cell cycle rate changed,the rate of G0/G1 phase of A549 cell was decreased,the rates of S and G2-M phase were increased,the apoptosis rate increased within time prolonged. PT-PCR showed that the killing effect of BPE on A549 cells wasn＇t the way of bax/bcl-2 gene. Conclusion BPE can inhibit the growth of A549 cell,and induce the apoptosis of the A549 cell.
