摘 要:目的观察早期糖尿病肾病大鼠肾脏线粒体功能和结构变化。方法将20只健康雄性大鼠随机分为正常对照组和糖尿病组,每组10只。糖尿病组大鼠采用链脲佐菌素诱导糖尿病模型。第10周末测定2组血糖、肾重/体质量、24h尿蛋白定量;检测各组肾脏线粒体丙二醛(MDA)、一氧化氮(NO)含量及锰超氧化物歧化酶(Mn-S0D)、总一氧化氮合酶(T-NOS)、组分ATP酶活性和线粒体呼吸控制率(RCR),分析线粒体比表面。结果与正常对照组比较,糖尿病组血糖、肾重/体质量、24 h尿蛋白定量明显增加(P<0.01),肾脏线粒体MDA、NO含量、T-NOS明显增加(P<0.01),Mn-S0D、组分ATP酶活性、线粒体RCR及比表面明显减少(P<0.01)。结论早期糖尿病肾病大鼠肾脏线粒体存在明显的氧化损伤和功能、结构障碍。Objective To explore renel mitochondrial functure and ultrastructural in the early diabetic nephropathy rats.Methods The diabetic model(DM) induced by streptozotocin(STZ) was adopted.Twenty male wistar rats were randomly divided into normal control(NC) group and DM group,with ten rats in each group.Ten weeks later,blood glucose level,the ratio of kidney weight/body mass(KW/BW),24h urinary protein excretion,the levels of MDA,NO,MnSOD,T-NOS,Na+,K+-ATPase,Ca2+-ATPase and respiratory function in the renal mitochondrial were measured repectively.Moreover,the ultrastructure of renal mitochondria were examined under transmission electron microscope(TEM).Results Blood glucose level,KW/BW,24 hours urinary protein excretion and the levels of MDA,NO,T-NOS in diabetic rats were significantly increased compared with NC group(all P0.01).The levels of MnSOD,Na+,K+-ATPase,Ca2+-ATPase and respiratory control rate,Speciaic surface in diabetic rats were significantly decreased than that in NC group(all P0.01).Conclusion This study suggested the renel mitochondria in the early diabetic nephropathy rats undergo a severe oxidative injure and damage of function and structure.
