硼替佐米诱导前列腺癌细胞株PC-3细胞凋亡的研究

摘　　要：目的探讨硼替佐米诱导前列腺癌细胞株PC-3细胞的凋亡作用。方法采用噻唑蓝(MTT)比色法观察2.6、10.4、41.6、166.4、260、665.6、1040、2662.4及10 649.6 nmol/L浓度硼替佐米对PC-3细胞的生长抑制作用,采用Annexin-V标记检测细胞凋亡情况;采用RT-PCR方法检测665.6 nmol/L硼替佐米作用PC-3细胞0、6、24及48 h bax、bcl-2和bim基因表达的变化。结果 2.6 nmol/L硼替佐米对PC-3细胞就有抑制作用,并呈时间和剂量依赖性抑制。665.6 nmol/L硼替佐米作用PC-3细胞24 h后,可见明显细胞核凝聚、固缩和碎裂;Annexin-V阳性表达率为18.5%,明显高于对照组(2.04%)。RT-PCR检测显示665.6nmol/L硼替佐米作用PC-3细胞24 h bax表达增强,而bcl-2和bim表达减弱。讨论硼替佐米可以抑制PC-3细胞生长,并通过上调bax表达,下调bcl-2和bim表达来诱导凋亡。ObjectiveTo explore the effect of Bortezomib on proliferation and apoptosis of prostatic carcinoma PC-3 cells.Methods MTT assay was used to detect the proliferation,while Annexin-V staining was used to investigate apoptosis of PC-3 cells.RT-PCR was used to analyze the mRNA expressions of bcl-2,bax and bim.Results Bortezomib inhibited the proliferation of PC-3 cells in time-and dose-dependent manners.After having treated with Bortezomib for 24 hours,PC-3 cells showed obvious nuclear condensation,chromatin condensation and fragmentation.Annexin-V-positive rate was 18.5%,significantly higher than that of the control group（2.04%）.RT-PCR showed that Bortezomib up-regulated the mRNA expression of bax in 24 h,while down-regulated expression of bcl-2 and bim.Conclusion Bortezomib inhibits the proliferation of PC-3 and induces apoptosis by up-regulating bax and down-regulating bcl-2 and bim.
