摘  要:目的探讨火棘多糖对小鼠肝、肾、脑和心各组织体内外的抗氧化作用。方法将昆明种小鼠分为正常对照组、模型组和火棘多糖高、中、低保护组。饲养8d后,正常对照组腹腔注射植物油溶液,其余各组腹腔注射0.15%CCl4植物油溶液。24h后眼球取血,分离血清,测定ALT和AST。用PBS制备肝、心、肾和脑匀浆;在组织匀浆中加入FeSO4和H2O2,诱导自由基的生成,测定各组织体内外的抗氧化作用。结果不同浓度的火棘多糖保护组均可降低血清ALT和AST的含量(P<0.01或P<0.05),降低各组织MDA的含量(P<0.01或P<0.05)。体外抗氧化实验能抑制脂质过氧化物的生成(P<0.01或P<0.05)。结论火棘多糖能降低CCl4肝损伤小鼠血清ALT和AST活性,对小鼠肝、肾、心和脑各组织体内外均有抗氧化作用,尤其对心、肾和肝组织的抗氧化作用较强。【Objective】To investigate the anti-oxidant effect in vitro and in vivo of Pyracantha fortuneana polysaccharides on mouse liver,kidney,brain,heart tissues.【Methods】Kunming mice were divided into normal control group,model group,Pyracantha fortuneana polysaccharides high,medium and low protection group which were fed 8 days.At the end of the feeding,the normal control group was intra-peritoneal injected with vegetable oil solution,and the rest of the group was intra-peritoneal injected with 0.15% CCl4 vegetable oil solution.After 24 hours,ALT and AST were determined through the separation of the eye blood serum.The liver,heart,kidney,brain homogenate were prepared with PBS;FeSO4 and H2O2 were added to induce the generation of free radicals,and the anti-oxidant action of the tissues in vitro and in vivo were tested.【Results】Different concentrations of Pyracantha fortuneana polysaccharides can reduce ALT and AST levels of the protect groups in serum (P 0.01 or P 0.05),lower MDA content organizations in various organizations (P 0.01 or P 0.05).Anti-oxidation in vitro experiments can inhibit the generation of lipid peroxides (P 0.01 or P 0.05).【Conclusion】Pyracantha fortuneana polysaccharides can reduce the activity of ALT and AST in serum on mouse's liver damaged by CCl4.Besides,it is indicated that the polysaccharides show antioxidation on liver,kidney,heart and brain tissues in vivo and in vitro,especially on the heart,kidney and liver.
