胸苷酸合成酶在大肠癌组织中的表达及其临床意义

摘　　要：目的探讨结直肠癌患者胸苷磷酸化酶(TS)表达水平与5-FU疗效及临床病理的相关性。方法对58例结直肠癌患者,采用免疫组织化学方法检测癌组织和正常结直肠粘膜组织中的TS表达。评价患者TS表达情况与5-FU化疗疗效、临床病理因素之间的相关性。结果 TS免疫组化结果为癌组织中TS表达明显高于正常结直肠粘膜组织,分别为55%(32/58)和14%(8/58).肿瘤进展时间(TTP)TS(-)者明显优于TS(+)者,分别为(6.5±1.9个月)和(3.8±1.2个月),具有统计学意义(P<0.05)。结论 TS检测对预测5-FU化疗疗效、判断预后有一定的指导意义,为结直肠癌的个体化治疗提供了理论依据。Objective To examine the expression level of thymidylate synthase（TS） in colorectal cancer;To investigate the correlations of TS to the sensitivity of fluoropyrimidine-based chemotherapy and clinicalpathological factors.Methods To fifty-eight colorectal cancer Patients,we used the immunohistochemical staining to check the expression of TS.To investigate the correlations of TS to the sensitivity of 5-FU chemotherapy and clinicalpathological factors.Results The expression level of TS in colon carcinoma was higher than that in normal colon tissues,the expression was 55%（32/58） and 14%（8/58） respectively.Correlation to histopathological parameters revealed a significant association between tumor dukes stage and the TS level.The time to tumor progression（TTP） of TS（-） patients and TP（＋） patients was 6.5±1.9 months and 3.8±1.2 months respectively.Conclusion The expression of thymidylate synthase（TS） is useful to predict the sensitivity of fluoropyrimidine-based chemotherapy and the prognosis of colorectal cancer.
