子宫内膜样腺癌中GELSOLIN和PTEN蛋白的表达及其临床意义

摘　　要：目的探讨子宫内膜样腺癌发生、发展的分子生物学机制,为子宫内膜样腺癌的生物学特性的评定及靶向基因治疗,寻找新的分子生物学指标。方法采用免疫组化SP法对100例不同子宫内膜组织进行两蛋白的检测,采用秩和检验及Spearman等级相关分析进行统计学处理。结果 GELSOLIN的表达情况N组优于A组及E组(P<0.05);无淋巴结转移者优于有淋巴结转移者(P<0.05)、浸润深度<1/2肌层者优于≥1/2肌层者(P<0.05)。PTEN的表达情况N组优于A组及E组(P<0.05);G1期优于G2+G3期、浸润深度<1/2肌层者优于≥1/2肌层者(P<0.05)。GELSOLIN与PTEN在肿瘤中的表达无相关性(P>0.05)。结论 GELSOLIN或PTEN表达降低能促进子宫内膜样腺癌的发生、发展,两蛋白的作用通路可能无交叉,互不影响。Objective To study the molecular mechanisms of development and progression of endometrioid adenocarcinoma and to identify new molecular markers for the evaluation of molecular properties and targeted gene therapy of endometrioid adenocarcinoma.Methods SP immunohistochemistry was used to examine Gelsolin and PTEN in endometrial tissues（n= 100）.Log-rank test and Spearman′s rank test were performed.Results The expression of Gelsolin was higher in group N than in group A and group E（P0.05）.That of Gelsolin was higher in patients without lymph node metastasis than in those with（P0.05）,and higher in patients with invasion depth 1/2 of muscular layer than in those with ≥1/2（P0.05）.The expression of PTEN was higher in group N than in group A and group E（P0.05）.That of PTEN was higher in patients with G1 and invasion depth 1/2 of muscular layer than in those with G2＋G3 and invasion depth1/2（P0.05）.Conclusion The downregulation of Gelsolin or PTEN could promote the development and progression of endometrioid adenocarcinoma in an independent manner.
