摘  要:目的比较量子点与异硫氰酸荧光素(FITC)标记检测不同分级和分期的前列腺癌组织中前列腺特异抗原(PSA)的表达,探讨量子点作为新型荧光探针的应用价值。方法对72例前列腺癌组织标本分别进行量子点和FITC荧光标记,结合肿瘤的分级分期对两种方法的荧光标记结果进行比较研究,并对荧光强度的衰减进行观察记录。结果两种标记方法对前列腺癌组织中PSA随肿瘤分级和分期的表达变化的显示具有一致性。连续激发下,FITC荧光在6min内急剧衰减,而量子点荧光在至少10d内可保持稳定,在1个月时仍有82%的阳性标本的荧光可以辨识。结论量子点荧光标记在生物医学实际应用中具有良好的敏感性和稳定性。Objective To compare quantum clots and fluorescein isothioeyanate (FITC) fluores- cence labeling for detection of prostate specific antigen (PSA) protein expression in prostate cancer correlated with different tumor stage and grade, and to investigate the value of quantum dots as a new type of fluorescent markers for biomedical applications. Methods Seventy-two eases of prostate cancer were divided into 3 groups ( A, B and C ) according to Gleason score, which corresponding to Gleason 2-4 points ( 19 cases) , 5-7 points (28 eases) and 8-10 points (25 cases). Quantum dots and FITC fluorescence labeling were performed to analyze the expression of PSA protein on the entire specimens, respectively. The intensity of fluorescence was interpreted immediately after labeling, and the fluorescence intensity decay was observed and recorded at 4 time points in within one month. Results The results of quantum dots and FITC fluores- cence labeling were consistent and demonstrated that the decreases in the positive rate and intensity of PSA expression accompanied with increased Gleason score and clinical stage of the tissue samples. The FITC fluo- rescence under continuous illumination faded quickly and became undeteetable after 6 min, however, only one ease with the fluorescence intensity change within 10 days for quantum dots fluorescence. Moreover, even to one month, the quantum dots fluorescence still could be seen in all except 9 specimens. Conclusion Fluorescent quantum dots have good sensitivity and stability for practical applications in biomedicine.
