摘  要:本文通过对上海市疾病预防控制中心收集并保存的、来源于上海崇明岛地区痰培养阳性肺结核患者的135株结核分枝杆菌北京基因型菌株进行可变数目串联重复序列(VNTR)和单核苷酸多态性(SNP)基因分型,并构建最小生成树(MST),来寻找适合研究北京基因型菌株微进化的VNTR组合及区分不同亚型的分子标志。结果显示,16位点VNTR与SNP的分型结果最匹配,适合于研究该地区北京基因型菌株的微进化;SNP单倍型主要集中在ST10(42.2%)、ST19(30.4%)、ST22(14.1%)3个类型;4个VNTR位点(Mtub21、QUB26、MIRU16和QUB4156)可能是区分北京基因型菌株不同亚型的分子标志。To find the variable number of tandem repeat (VNTR) locus combinations that adapt to study the microevolution of Mycobacterium tuberculosis (M. tuberculosis) strain of Beijing genotype and the molecular markers to distinguish different sublineages,VNTR and single nucleotide polymorphism (SNP) genotyping were used to genotype 135 strains from culture-positive sputum of tuberculosis patients (collected and preserved by Shanghai Municipal Center for Disease Control and Prevention) in Chongming Island,Shanghai. Based on the data,the minimum spanning tree (MST) was constructed. The results showed that the genotyping information of 16-locus VNTR best matched the SNP genotyping data,indicating that it was applicable in the microevolution study of M. tuberculosis strain of Beijing genotype in Chongming Island,Shanghai. SNP haplotypes were mainly concentrated in ST10 (42.2%),ST19 (30.4%),and ST22 (14.1%). Four VNTR loci (Mtub21,QUB26,MIRU16 and QUB4156) may be the molecular markers to distinguish different sublineages of Beijing genotype strains.
