摘  要:目的:探讨熊果酸(ursolic acid,UA)对人腺样囊性癌ACC-83细胞增殖及凋亡的影响。方法:MTT检测UA对ACC-83细胞体外生长的抑制作用;流式细胞仪测定细胞的周期和凋亡;瑞氏-吉姆萨染色,显示凋亡细胞形态;Western印迹法检测Bcl-2、Bax两个重要的细胞凋亡相关基因和细胞凋亡相关特殊蛋白COX-2表达的变化。结果:UA以剂量和时间依赖的方式抑制ACC-83细胞生长,以剂量依赖的方式使ACC-83细胞周期阻滞在S期,诱导细胞凋亡;染色可见细胞呈明显的凋亡特征;Western印迹法测得UA下调ACC-83细胞的Bcl-2和COX2的蛋白表达,随着药物浓度增加逐渐减少,对Bax的表达并无明显改变。结论:UA能够抑制ACC-83细胞的增殖,并使细胞大量累积于S期,UA可能通过减少COX2和Bcl-2的表达、降低Bcl-2/Bax比例来诱导细胞凋亡。Objective:To assess the proliferation inhibition effects and apoptosis induction of ursolic acid on human ACC 83 cells.Methods:ACC-83 cell proliferation was measured with MTF assay.Cell cycle phase distribution and hypodiploid DNA were determined by flow cytometry.Morphological analysis were viewed with Wright-Giemsa stain.Two important apoptosis related genes Bcl-2 and Bax and the particular COX-2 protein were examined by Western-blot. Results:MTT assay showed that UA inhibited ACC-83 cell proliferation showed in a time- and dose-dependent manner. UA induced ACC-83 cell cycle arrested in S phase and the cell apoptosis characterized morphologically.COX-2 protein expression level decerased by UA in a dose-dependent manner,Bcl-2 portein expression decreased also in a dosedependent manner,but Bax expression remained unchanged.Conclusion:Ursolic acid can inhibit ACC-83 cell proliferation,and to a large number of cells accumulated in S phase,Ursolic acid may downregulate expression of COX-2 and Bcl-2 and reduce the Bcl-2/Bax ratio to induce apoptosis.
