摘  要:目的:研究不同温度的顺铂对人卵巢癌细胞株SKOV3的增殖抑制效应及S100A4 mRNA表达的影响。方法:以体外培养的人卵巢浆液性乳头状囊腺癌细胞株SK-OV3为研究对象,按不同的热疗温度,用四甲基偶氮唑蓝(MTT)法测定各组细胞抑制率;流式细胞仪(FCM)分析细胞周期分布;用半定量逆转录聚合酶链反应(RT-PCR)技术检测各组S100A4 mRNA的表达。结果:(1)各组细胞抑制率均较阴性对照组增加(P<0.05),43℃组作用最明显(P<0.01);(2)FCM分析细胞周期显示各组细胞与对照组比较,G0/G1期细胞比例增高,S期细胞比例下降(P<0.05),细胞生长阻滞于G0/G1期;(3)RT-PCR结果显示,不同温度作用后S100A4 mRNA的表达均低于阴性对照组(P<0.01),以43℃组表达最低。结论:不同温度的顺铂热化疗均可抑制卵巢癌细胞株SKOV3增殖及下调S100A4 mRNA的表达,从而抑制其侵袭转移,以43℃时作用最显著。Objective:To investigate the effect of cisplatin combined with hyperthermia in different temperature on the cell growth and expression of S100A4mRNA.Metheds:The cultured human ovarian carcinoma SKOV3 cells were treated with cisplatin at different temperatures(37℃,39℃,40℃,41℃,42℃,43℃,44℃,45℃).MTT assay was used to determine the inhibition rate of cell growth.Flow cytometry(FCM) was used to analyse changes of cell cycle.Reverse transcription polymerase chain reaction(RT-RCR) technique was used to observe the expression of S100A4 mRNA.Results:Compared with control group,the inhibition rate of cells in experimental groups increased(P0.05),while the expression of S100A4 mRNA decreased significantly(P0.01).When the temperature reached 43℃,the inhibition rate increased to the maximum but the expression of S100A4 mRNA decreased to the minimum.Cell cycle analysis by FCM indicated the increase of the proportion of cells in G0/G1 phase and decrease of S phase,the cell cycle could be blocked gradually at G0/G1 phase by hyperthermia(P0.05).Conclusion:Cisplatin can inhibit the growth of SKOV3 cells in vitro and decrease the expression of S100A4 mRNA at different temperature,with 43℃ affects most apparently.
