摘 要:目的研究食管鳞状上皮细胞癌(简称食管鳞癌)组织中脾酪氨酸激酶(Syk)及p53基因的表达及二者的关系,并探讨其与食管鳞癌发生及发展的相关性。方法用快捷免疫组化法及RT-PCR法检测食管鳞癌组织及其对应癌旁组织中Syk蛋白及mRNA的表达,同时检测p53蛋白及mRNA的表达,分析食管鳞癌中上述2个基因表达变化的相关性,并观察其临床病理意义。结果食管鳞癌组织Syk蛋白及mRNA的表达显著低于癌旁组织(P<0.05);食管鳞癌组织p53蛋白及mRNA的表达显著高于癌旁非肿瘤组织(P<0.05);Syk蛋白表达与p53蛋白(突变型)表达呈负相关(rs=-0.604,P<0.05);Syk蛋白表达与食管鳞癌TNM分期、淋巴结转移情况相关(P<0.05),与肿瘤大小无关(P>0.05)。结论 Syk在食管鳞癌中表达减弱及缺失与其发生和转移倾向相关;Syk可能是食管鳞癌的肿瘤抑制基因,其作用可能是p53(野生型)依赖性的。Objective To investigate the expressions of Syk and p53 gene in human esophageal squamous cell carcinoma and matched-adjacent tissues,and then explore their correlation with the genesis and progression of esophageal squamous cell carcinoma.Methods The expressions of Syk and p53 protein and mRNA in tumor specimens and adjacent non-tumor specimens of esophageal squamous cell carcinoma were assayed by rapid immunohistochemistry and RT-PCR method,and their correlations with patients' clinical biological behaviors were analyzed at the same time.Results The positive expressions of Syk protein and mRNA in esophageal squamous cell carcinoma specimens were significantly lower than those in adjacent non-tumor specimens(P0.05).The positive expressions of p53 protein and mRNA in esophageal squamous cell carcinoma specimens were significant higher than those in adjacent non-tumor specimens(P0.05).The expressions of Syk protein and p53 protein(mutant type) were negatively correlated with each other(rs=-0.604,P0.05).The expression of Syk protein was related to tumor TNM stage and lymph node metastasis(P0.05),but not related to tumor size(P0.05).Conclusion The reduction and silence of Syk expression in esophageal squamous cell carcinoma may be associated with its malignant metastatic tendency;Syk gene may play the role of tumor-suppressing gene in esophageal squamous cell carcinoma genesis,which may be p53(wild type) gene dependent.
