摘 要:目的探讨高脂饮食对SD大鼠心脏功能的影响以及吡格列酮干预的效果。方法 36只8周龄雄性SD大鼠随机分为3组:正常饲料组(NC组,12只),高脂饲料组(HF组,12只)及吡格列酮干预组(HP组,12只)。饲养28周后:①检测血游离脂肪酸(FFA)、甘油三酯(TG)及心肌组织TG含量;②计算左心室质量指数;③正常血糖高胰岛素钳夹试验;④观察大鼠高频超声心动图;⑤采用多导生理记录仪活体检测大鼠左心室最大收缩速率(+dp/dtm ax)和左心室最大舒张速率(-dp/dtm ax)。结果①HF组大鼠血FFA、TG及心肌TG含量明显增高(P<0.01),吡格列酮干预后,以上指标明显改善(P<0.01);②HF组和HP组左心室质量指数明显增高(P<0.01);③HF组G IR较NC组显著降低(P<0.01),HP组G IR明显改善(P<0.01);④HF和HP组左心室后壁收缩末期厚度(Ts)、左心室后壁舒张末期厚度(Td)均明显增加(P<0.01);HF组射血分数(EF)、短轴缩短率(FS)和心指数(C I)降低(P<0.01),吡格列酮干预后EF、FS和C I升高(P<0.01);3组室壁收缩增厚率(△T%)、心率(HR)差别无统计学意义(P>0.05);⑤HF组心脏+dp/dtm ax和-dp/dtm ax均明显下降(P<0.01);吡格列酮干预后+dp/dtm ax和-dp/dtm ax均较HF组改善(P<0.01)。结论高脂饲养可导致大鼠心脏功能受损,吡格列酮干预后可以明显改善大鼠心脏功能,其机制与改善外周胰岛素抵抗有关。Objective To investigate the effects of high-fat diet and pioglitazone intervention on cardiac function of SD rats.Methods Thirty-six 8-week-old male Sprague-Dawley rats were randomly divided into three groups,including a normal diet(NC) group,a high-fat diet(HF) group,and a pioglitazone-treated(HP) group.After feeding for 28 weeks,(1) the concentrations of free fatty acids(FFA),triglyceride(TG),and myocardial TG in fasting serum were measured;(2) left ventricle weight index(LVMI) was calculated;(3) hyperinsulinemic-euglycemic clamp technique was performed;(4) echocardiographic studies were performed;(5) the maximum velocity of myocardial contraction(+dp/dtmax) and the maximum velocity of myocardial diastole(-dp/dtmax) of left ventricle were measured using a physiological polygraph.Results (1) The concentrations of serum FFA,TG,and myocardial TG in the HF group were remarkably higher than those in the NC group(P0.01),while these values were remarkably lower in the HP group than in the HF group(P0.01);(2) the LVMIs in the HF group and HP group increased significantly(P0.01);(3) the glucose infusion rate(GIR) was significantly lower in the HF group than in the NC group(P0.01),while it was substantially higher in the HP group than in the HF group(P0.01);(4) the end-systolic thickness and end-diastolic thickness of left ventricular posterior wall increased significantly in the HF group and HP group(P0.01);the ejection fraction(EF),fractional shortening(FS),and cardiac index(CI) were lower in the HF group than in the NC group(P0.01),but they were higher in the HP group than in the HF group(P0.01).The wall systolic thickening rate(△T%) and heart rate(HR) had no statistical differences among the three groups(P0.05);(5) +dp/dtmax and-dp/dtmax were significantly lower in the HF group than in the NC group(P0.01),while they were higher in the HP group than in the HF group(P0.01).C
