摘 要:目的:观察温热处理对K562细胞蛋白质表达谱的影响。方法:将培养获得的K562细胞在41℃恒温处理1h,以37℃作为对照,采用荧光双向凝胶电泳技术分离全细胞蛋白质,分子成像仪获取图像后软件分析差异表达的蛋白质点。结果:温热处理K562细胞与对照组细胞蛋白质表达谱分析比较差异表达蛋白质点数为65个,其中温热处理后表达上调的蛋白质点27个,下调的蛋白质点38个。结论:研究将为阐明热疗的分子基础提供可靠的实验依据,随着研究的深入必将推动基于热疗的肿瘤生物治疗的更大发展和广泛的临床应用。Objective To investigate the effects of lukewarm treatment on the expressing spectrum of protein in K562 cells. Methods As compared with those treated under 37℃,cultured K562 cells were treated under 41℃ ior 1 hour,then whole cellular proteins were separated by fluorescent 2-DE.Images obtained by molecular imaging were analyzed by software to discover different expression protein spots. Results 65 protein spots were differentiall? expressed between K562 eells in the two groups, among which, 27 spots were up-regulated and 38 spots were down-regulated in K562 cells after lukewarm treatment. Conclusion This study paside ide the experimental foundation fur elucidating molecular mechanism of thermotherapy.
