摘　　要：目的：建立血清Ⅳ型胶原蛋白浓度测定的酶联免疫试验方法，辅助诊断肝硬化。方法：制备过氧化物酶－兔抗人ＣＯＬⅣ多抗结合物，用棋盘滴定试验确定包被抗体和标记抗体浓度。进行检出限，精密度，回收率，特异性，灵敏度和标准曲线的制备；初步用于７９例下落空和３４例肝纤维化患者。Objective: To develop an experimental way of enzyme linkded immunoassay for the density measurement of serum tpe Ⅳ collagen. Methods: HRP rabbit anti - human type Ⅳ collagen PcAB was prepared as labelled antibody and the optimum concentration of labelled antibody and coationg buffuer were detected with chessboard test; the lower limits of detection ,precision ,recovery. specificity, sensitivity were determined so as to establish the standan curve used in 79 normal and 34 liver patients with cirrhosis. Result: The average intra - assay CV was 6 .098% and the inter - assay CV was 4. 30% ,the recovery of high and low concertration of type Ⅳ collagen was 95% and 98%. With this assay,the average concertration of healthy donors type Ⅳ collagen was 67. 6 ±24. 6ug/L, the average value of liver cirrhosis' type Ⅳ collagen was 215. 96 ±99. 0ug/L. Conclusion: ELISA is simple, precise. specific and lower cost, so it can be used in the diagnosis of liver cirrhosis.
