鼠脑胶质瘤模型的建立与增强磁共振成像研究

摘　　要：目的：应用ＭＲＩ 技术观察脑胶质瘤动物模型中肿瘤的生长和ＭＲＩ 表现。使用不同ＭＲＩ 增强方式，探讨腹腔给药增强的可行性和变化规律。材料和方法：１． 选用Ｃ６ 大鼠脑胶质瘤细胞株。制成细胞悬液，接种于ＳＤ 雄性大鼠。２． 使用超导中场磁共振成像系统。常规ＳＥ 序列。增强检查使用国产磁影萄胺。使用膝关节线圈。增强检查运用２ 种方法，经腹腔给药和经尾静脉给药。３． 观察：（１） 肿瘤不同生长期，大小变化情况。（２） 观察肿瘤区的ＭＲＩ信号变化。（３） 观察不同给药方式和给药剂量，肿瘤强化的情况。结果：１ 、Ｔ２ 加权正常脑组织、肿瘤组织和周围水肿组织间的信号差异最大。２ 、经腹腔内注入Ｇｄ —ＤＴＰＡ，理想的给药浓度为０ ．２５ ｍ ｍ ｏｌ／Ｋｇ 。３ 、经静脉和腹腔注射Ｇｄ —ＤＴＰＡ 后肿瘤的显示情况比较，均可良好显示肿瘤。浓度为０ ．２５ｍ ｍ ｏｌ／Ｋｇ 时，肿瘤的强化率的比较，均得到满意的结果。结论：ＭＲＩ 的增强技术可精确地了解鼠脑胶质瘤的生长变化情况和肿瘤的形态学改变。腹腔给药增强方式，大大改善和简化了ＭＲＩ 增强检查过程，有效地提高了工作效率。Purpose:Surveying tumor growth and MRI manifestations in glioma of rat brain model and the evaluation of the growth of rat glioma with the injection of Gd-DTPA intra peritoneally with the different injection methods of Gd-DTPA for comparison.Materials and Methods:1、The model of rat cerebral glioma were extablished by stereotactic inoculating C6 cells into Sprague-Dawley rat in date nucleus.2、The enhancements of tumors were evaluated with different types of the injection of Gd-DTPA,intra peritoneally and inteaveronely.3、MRI menifastations of tumour were studied in different periods including the signals、sizes and shapes of the tumor.Results:1、There were high signal differences among normal tissues、tumor tissues and edemal tone tissues at T2 weighted images.2、The appropriate dosage of Gd-DTPA injected into peritoneun is 0.25mmol/Kg.3、The satisfied observations of the growth of rat glioma all can be obtained with different injection methods of Gd-DTPA.Conclusion:MRI enhancement is a good method to observe rat glioma and the injection of Gd-DTPA intra peritoneally is consided as the conventional examination because of its convenience.
